Résumé:
The current speed at which novel high-throughput technologies are developed and large-scale biological data are produced offers tremendous opportunities for enhancing our molecular understanding of biological systems. One of the biggest challenges will be to interpret these large-scale biological “omics” data and to develop computational and statistical approaches to generate a coherent biological model that will enable us to address novel questions about the consequences of genetic and environmental perturbations in complex biological systems.

We are delighted to be holding this biannual event again after very successful meetings in 2011, 2013, and 2015, where renowned plant researchers gave inspiring talks and where many discussed their latest research in the field. However, unlike the previous three meetings, which were held in Amsterdam, the fourth PGE meeting will be held in Sitges (Barcelona), Spain, 1-3 October 2017.

EMBL Course: Whole Transcriptome Data Analysis

The course participants will use BioConductor software and Illumina BaseSpace Cloud apps. R-coding skills and deep knowledge of statistics are not necessary for attendance because most analyses will be performed with OneChannelGUI (a graphical interface to a series of BioConductor tools, which enables handling microarray data analysis as well as RNA-seq data without requiring programming skills). Computers for hands-on exercises will be provided along with demo data sets. Upon request, participants could bring along their own data to analyse.

This conference will provide an opportunity to learn about and keep up to date with the rapidly progressing area of cancer genomics. It will cover presentations from cancer genome projects, the areas of cancer functional genomics, systems biology, cancer mouse models and the translation and clinical impact of scientific results obtained. The meeting will bring together leading scientists from across these areas for a unique opportunity to interact and stimulate further integration of these efforts.

European Cancer Genomics research is particularly strong in the Heidelberg area, with three International Cancer Genome Consortium (www.icgc.org) projects being currently pursued with significant participation from Heidelberg institutions, including EMBL. As in our 2015 meeting, a large number of renowned speakers will be invited, with a strong representation of overseas speakers. This conference brings together an international group of scientists to present and discuss the emergence of and new developments in epigenomics/epitranscriptomics and their impacts on basic biology as well as human diseases, with a focus on DNA and RNA methylation. In addition, the conference provides hands-on workshops for junior and established scientists on key sequencing and computational methodologies and resources for epigenomics/epitranscriptomics research. These components will ultimately catalyze DNA and RNA methylation research and accelerate the pace of discovery. Obesity and type 2 diabetes are major drivers of chronic disease and premature death. A promising strategy to reduce obesity and improve metabolic health is through promoting energy expenditure via thermogenesis, including obligatory (accompanies all metabolic processes) and facultative (adaptive) mechanisms. Obesity and insulin resistance also lead to profound metabolic changes in various cells/tissues that contribute to tissue dysfunction. This conference brings together basic and clinical researchers from industry and academia focused on the study of cellular and organismal energy metabolism in the context of metabolic disease. A variety of approaches and topics will be featured, including: basic mechanisms of mitochondrial and cellular energetics; the emerging role of brown and beige fat in adaptive thermogenesis; central regulation of energy expenditure; and the effects of exercise on muscle and whole-body metabolism. In addition to the plenary sessions, the meeting will feature workshops on emerging research areas and novel therapeutic opportunities. Overall, the goals of the conference are to: 1) Provide a cutting-edge understanding of the basic mechanisms that control cellular and whole-body energetics; 2) Discuss the physiological role of brown fat and other energy expenditure pathways in humans; 3) Consider novel strategies for increasing energy expenditure and combating metabolic disease; and 4) Engage attendees in critical discussions about important areas for future research.

Keystone Symposia: Precision Genome Editing with Programmable Nucleases

Genome editing refers to the use of programmable DNA-targeting platforms to manipulate genetic and epigenetic information in living cells and organisms. Genome editing has been recently democratized by the development of RNA-guided endonucleases repurposed from CRISPR-Cas microbial defense systems against viruses or plasmids. Whereas novel tools such as Cpf1 and RNA-guided deaminases are being developed to expand the scope of genome editing, old tools such as ZFNs and TALENs are likely to undergo innovations as well. These tools enable genome-wide genetic screens in cell lines and genetic studies in both model and non-model organisms. Furthermore, genome editing holds great promise for broad applications in medicine and biotechnology, raising public interest and concerns. Yet genome editing is still limited by many factors such as inefficient delivery in vivo, off-target effects, and unwanted, alternative DNA repair. This conference offers opportunities to learn about new developments, advancements and applications in the field.

Keystone Symposia - Cancer Epigenetics: New Mechanisms, New Therapies

The field of epigenetics has recently emerged as an essential component of oncology research, with altered epigenetic modifications considered to be critical hallmarks of cancers. Unlike mutations, epigenetic aberrations are reversible and as a result, epigenetic therapies hold great promise as anti-cancer agents. For these novel therapies to succeed, a key challenge is to fully understand the nature and the mechanisms that lead to epigenetic changes in cancer. Another essential challenge is to make sense of the avalanche of data flowing from increasingly powerful "omics-based" technologies. Despite these challenges, never has the potential for scientific and clinical breakthroughs in this field been greater. Bringing together both the academic and the pharmaceutical communities, this Keystone Symposia conference will focus on: 1) The newest findings concerning the mechanisms by which epigenetic pathways are established and maintained in normal and stem cells, and how these go awry during aging and cancer, with a special emphasis on histone modifications, chromatin remodeling and the unexpected increased number of DNA modifications ((hydroxy)methylcytosine and methyladenine); 2) The growing roles of noncoding RNAs in cancer and the emergence of the new and exciting realm of RNA epigenetics; 3) Use of new omics technologies to interrogate altered epigenetic landscapes in tumors and assess their clinical implications; and 4) Novel and promising therapeutic drugs aimed at reversing specific epigenetic alterations. Ultimately, the meeting will provide great opportunities for senior and junior scientists to meet the leaders in the field from both academia and industry, hence forging exciting interdisciplinary interactions and creating potential new collaborations in the booming area of cancer epigenetics.

The main objective of this conference is to bring people together from diverse disciplines to exchange ideas, promote cross-disciplinary collaborations and to form a synthesis of appropriate systems-level approaches. The meeting is therefore purposely broad to cover all aspects of genomics to systems biology, a unique combination that is highly appreciated by the participants. Over the past decade, this EMBO conference has therefore served as an important venue in helping to shape the field, or to be more precise to help generate a community of scientists that come from very diverse disciplines, each with the common goal to understand the systems level properties of their system of interest.
