 Zusammenfassung Fragile X syndrome (FXS) is the most common form of inherited intellectual deficiency and the most common identifiable single gene cause of autism, affecting 1 in 5,000 males and a lesser number of females. Autism spectrum disorders (ASD) occur in up to 2/3 of males and 1/3 of females with FXS. The Fragile X gene (FMR1) was cloned in 1991 and since then a large field has grown with roughly one hundred labs using techniques from biochemistry through genetics to model organisms to elucidate the functions of the FMR1 protein (FMRP). FMR1 gene homologs have been found in Drosophila and mice, and useful null mutant models generated in both. FMRP has been found to be involved in the regulation of specific messenger RNA transport, localization and expression in neurons. Briefly put, FMRP is thought to bind and help transport a crucial subset of messages to the sites of neural action (the synapses). At those synapses, FMRP is a critical switch that mediates changes in local protein expression in response to neural activity - in effect causing the synapses to strengthen or weaken as required in response to experience. When FMRP is reduced or missing in an affected person, the synapses are much less able to change with experience and learning is greatly reduced. More recently, specific signaling mechanisms have been proposed to mediate this control at the synapse, via a specific set of Gq-linked (neurotransmitter) receptors, including group I mGluRs (metabotropic glutamate receptors). It has become clear that in addition to clinical overlap between FXS and ASD, there is likely substantial overlap in the molecular pathology of the two disorders. As such, molecular defects known to cause ASD may involve other proteins in the signaling pathways that are regulated by or regulate FMRP activity, may involve proteins for which synaptic translation is regulated by FMRP, or may involve defects in neurotransmitter systems shown to be dysregulated in FXS models. Molecules aimed at targets in pathways that are dysregulated in the absence of FMRP are now being developed and tested in academic laboratories and through the pharmaceutical industry, in order to offer effective drug therapies for affected patients with FXS. It is expected that many of these targeted treatments will have therapeutic overlap in subsets of individuals with ASD.

This conference will bring together leading scientists and clinicians in the Fragile X and ASD fields. Topics will include molecular genetics, model system characterizations, synaptic signaling and function studies, small molecule trials and clinical reports.

 Zusammenfassung The regulated trafficking, morphology and function of mitochondria is essential for providing ATP at the correct spatial location to power neural computation. Mitochondria also sequester and buffer Ca2+, and play a key role in apoptotic signaling, so their positioning and function also affects regulation of Ca2+ dynamics and neuronal death. In neurons, the concentration of mitochondria in specific regions such as growth cones and synapses is important for correct neuronal function and development. Moreover mutations in proteins regulating mitochondrial dynamics compromise neuronal development and the formation, function and plasticity of synapses. Consequently, defective mitochondrial trafficking and function is increasingly implicated in neurological diseases. This meeting will focus on the latest insights into our understanding of the mechanisms that control mitochondrial trafficking, form and function in neurons. The latest approaches for imaging cellular bioenergetics and mitochondrial positioning will also be a focus. How altered mitochondrial dynamics contribute to neuronal dysfunction in neurological diseases including Alzheimer’s, Parkinson’s and Huntington’s diseases will also be a key focus.

 Zusammenfassung Neurodegenerative and neurodevelomental diseases represent a major scientific challenge for the biomedical research community and a growing public health problem. Advances in human genetics and the identification of causative genes have allowed the application of modern biological tools to study the disease mechanisms. As a result it has become increasingly clear that pathogenesis of these complex brain disorders are not as simple as previously thought. This GRC intends to bring together investigators who are at the forefront of the field to present and discuss their latest findings and concepts. The Conference will feature a wide range but related topics that are relevant to disease mechanisms, such as synaptic dysfunction, autophagy, protein misfolding and aggregation, mitochondrial biology, and RNA binding proteins. Invited speakers represent a wide variety of scientific disciplines, including genetics, biochemistry, cell biology, electrophysiology, imaging and systems biology. The Conference will also provide opportunities for junior scientists and graduate students to present their work in poster sessions and interact informally with leaders in the field. Over the last decade, dramatic progress has been made in molecular biology, biophysics and genetics that impact our understanding of brain energy metabolism, neural organization, cell signaling and vascular regulation. In addition, new technologies have emerged to measure blood flow and metabolism with high spatial and temporal resolution. The stage is set to use the methodological progress to address fundamental issues related to the organization of brain function, blood flow and metabolic activity. We anticipate that progress in this field will drive new discoveries in the experimental and clinical neurosciences and impact diagnosis and treatment of stroke and other neurodegenerative disorder. The biannual Gordon Research Conference is devoted to presentations of frontline research of brain metabolism, cell signaling, cell-cell interactions and vascular regulation in the normal and injured brain.  Zusammenfassung The 2012 Gordon Conference on Neural Development in Newport, Rhode Island, will bring together a unique and international group of scientists with the common research goal of exploring the molecular, cellular and functional basis of brain development. Invited speakers will present cutting-edge research on topics ranging from brain patterning, neural stem cells, cell fate specification, neuronal migration and polarity, synaptic development, circuit development to new insights into human brain development and evolution. Each talk will be followed by discussions in which students and postdocs are especially encouraged to participate. The meeting will highlight the diversity of experimental approaches and model systems (e.g. mammals, zebrafish, C. elegans, Drosophila) that have led to major breakthroughs in the field. Recent technical achievements in molecular genetics and in vivo imaging relevant to research in neural development will be discussed extensively. Poster sessions and informal gatherings will provide additional opportunities for informal interactions among junior and senior researchers, and foster collaborations and further discussion. Some short talks by participants will be selected from the poster abstracts.  Zusammenfassung The 2012 Gordon Conference on Mechanisms of Epilepsy and Neuronal Synchronization will present cutting edge research on the theme of functional reorganization in the epileptic brain. The issues will be addressed at multiple levels, from cellular and molecular to circuits and networks. The focus will be on three important and persistent questions in the epilepsy field. What are the mechanisms underlying acute seizures? What progressive changes in brain organization and function lead to the chronic condition of epilepsy? What are the relationships between the mechanisms of epilepsy and the co-morbidities that are frequently encountered in this disorder? These questions will be considered specifically in sessions devoted to epileptogenesis, seizure generation, and developmental epilepsies and their co-morbidities. The issues raised during these sessions, including the reorganization of neural circuits in epilepsy, will be further developed in sessions that focus on exciting new methods of circuit analysis and their application in epilepsy research; mechanisms associated with reorganization of interneurons and alterations in chloride homeostasis; and the reorganization of several ion channels that could suggest targets for therapy. The purpose of the Conference is to bring together specialists from multiple disciplines to discuss the complex issues that arise in the processes of seizure generation and progression. It is expected that the exchange of new ideas will influence the critical next stage of experimental epilepsy research.

The collegial atmosphere of the Conference, with programmed discussion sessions as well as opportunities for informal gatherings in the afternoons and evenings, will provide opportunities for scientists to brainstorm and promote increased interactions among investigators in diverse areas of basic epilepsy research. The Conference will provide opportunities for junior scientists and graduate students to present their work in poster format and exchange ideas with leaders in the field. In addition to presentations by invited speakers, 8 short talks will be selected from submitted abstracts one to two weeks prior to the start of the meeting.

With these new approaches, investigators can control neuronal activity or signaling with unprecedented specificity and precision. These methods are within the reach of most investigators, and are replacing less selective techniques of electrical stimulation, lesions or pharmacologic manipulations. These approaches are not only a boon to basic researchers, but also have substantial potential for development of novel clinical treatments for a range of nervous system disorders.
