The combination of next-generation sequencing and analysis as applied to experiments studying the cancer genome has revolutionized our understanding of cancer biology. Large-scale cancer genomics has further established a baseline that is now poised to effect the translation of cancer genomics into the clinical setting, effectively transforming patient care. The barriers and challenges to translation are significant, not the least of which involves the transition of research-grade computational and interpretational analysis to clinical-grade analysis. Assays must be highly refined, validated and accurate. How the use of genomic approaches to monitor patient response to therapy, the onset of acquired resistance to therapy and other aspects of patient care will be accepted as a more sensitive partner with conventional imaging approaches must be addressed. Appropriate assignment of therapies to specific gene/protein alterations also will be of paramount importance, especially as the numbers and types of targeted therapies increase. Genomics will also impact our understanding of the immune system's interaction with tumors, leading to the development of personalized vaccine strategies that may provide more durable responses.

Over the past decade, there has been a greater understanding of genomic complexity in eukaryotes ushered in by the immense technological advances in high-throughput sequencing of DNA and its corresponding RNA transcripts. This has resulted in the realization that beyond protein-coding genes, there are a large number of transcripts that do not encode for proteins and, therefore, may perform their function through RNA sequences and/or through secondary and tertiary structural determinants. This Keystone Symposia meeting is focused on the latest findings concerning different classes of noncoding RNAs that use different strategies to regulate transcription. These are exciting times for discovering the biological scope and the mechanism of action for these RNA molecules, which have roles in dosage compensation, imprinting, enhancer function and transcriptional regulation, with a consequently great impact in development and disease. Why point mutations in a general transcription factor are associated with specific forms of human malignancies has been a major question in cancer biology. Enhancers are DNA regulatory elements that can be transcribed and are key regulators of tissue-specific gene expression in development; their malfunction through point mutations in either regulatory elements or factors modulating enhancer-promoter communication could result in development problems. Indeed, recent genome-wide studies in the cataloging of somatic mutations in cancer have identified mutations in intergenic sequences encoding regulatory elements and in specific transcription factors that function in enhancer-promoter communication. This Keystone Symposia meeting will review recent reports in this regard and discuss the concept of enhancer malfunction in the misregulation of gene expression in development and in disease pathogenesis. It is now evident that an integral component of immune regulation is through the metabolic pathways necessary to support energetically demanding protective or pathogenic responses. Deciphering these links in the emerging field of immunometabolism is critical to understanding basic immune function. Activation, differentiation and function of M1 & M2 macrophages, Th1, Th2, Th17 & T regulatory T cells as well as effector and memory CD8 T cells are all inexorably linked to pathways that regulate cellular metabolism. Likewise, systemic metabolism and metabolic diseases are very much influenced by immunity, as the metabolic syndrome in obesity is mediated in part by dysregulated immune cells similar to inflammatory and autoimmune disease states. This meeting addresses how immune cells and functions rely upon and are controlled by specific metabolic pathways and nutrient availability and will bridge systemic and cellular metabolic regulation of immune function. Metabolic reprogramming in the activation of adaptive and innate immune cells and the regulation and role of specific metabolic programs in distinct lymphocyte or macrophage subsets will be addressed. The conference will also present new findings on signaling pathways and the influence of immune cell metabolism on anti-tumor immunity, autoimmunity and metabolic diseases. It is anticipated this meeting will be of broad interest to the immunology and metabolic communities. The focus of this meeting on how factors that influence immune cell metabolism impact a variety of inflammatory disorders will provide new fundamental insight into normal immune function as well as opportunities to treat immunological and metabolic diseases. The past decade has seen tremendous developments in novel cancer therapies, through targeting of tumor cell-intrinsic pathways whose activity is linked to genetic alterations, as well as the targeting of tumor cell-extrinsic factors such as growth factors. Remarkable clinical success of checkpoint inhibitors as well as adoptively transferred genetically engineered T cells further demonstrates the critical role of T cells in cancer control and rejection. However, many patients still do not respond to checkpoint inhibitor therapies, possibly due to the lack of T cell repertoire with specificity against cancer antigens. This creates the need for effective means of expanding the T cells in patients via immunization, i.e., cancer vaccines. Much progress has been made in recent years in this regard, and several phase III clinical trials testing various approaches to therapeutic vaccination are ongoing. The challenge for next-generation vaccines is to resolve the discrepancy between the immune and clinical efficacy measured by the rate of cancer rejection. The future immunotherapy of cancer lies in combination approaches targeting T cells as well as cancer genes to combat underlying inflammation. Spectacular progress has been made in these two parallel fields, i.e., cancer genomics and genetics and tumor immunology. It is time now to link these fields to enable the linking of genetic alterations with the type of immune response. The meeting will therefore host cancer geneticists, cancer biologists, experts in cancer antigen presentation, tumor immunologists and vaccinologists. It will discuss the immunological basis for therapeutic cancer vaccines and how the current understanding of cancer genomics, antigen presentation and T cell biology might enable development of next-generation curative therapies for patients with cancer. This meeting will be held jointly with  Antibodies as Drugs.

Cancer stem cells have now been recognized in a wide variety of human tumors. The existence of cancer stem cells poses both tremendous challenges and tantalizing potential for our ability to successfully combat cancer. This Keystone Symposia meeting aims to bring together scientists who are interested in better understanding the mechanisms by which cancer stem cells contribute to tumor heterogeneity, self-renewal and drug resistance. One area of emphasis will be intercellular communication. Cellular context and microenvironment establish the tumor niche and shape the interaction of the tumor with the immune system. In addition, developmental signal transduction pathways such as Hippo, Notch and Wnt, frequently deregulated in cancer, increasingly offer tangible opportunities for therapeutic intervention. Another important area of emphasis will be efforts to better understand the mechanisms and importance of tumor heterogeneity. Techniques such as clonal analysis and barcoding are shedding important light on the interplay between tumor evolution, differentiation, miRNA control and epigenetics. This is an era of rapid progress in understanding cancer stem cell biology with great potential to benefit human health.

The local and systemic tumor microenvironments (TME) play a pivotal role in all stages of cancer development, including initiation, malignant conversion and metastasis, as well as regulating how neoplastic cells respond to cytotoxic, targeted and immune-based therapies. Research over the last decade has revealed the incredible diversity and plasticity that constitute the TME. Thus, the focus of this meeting will be to bring together a wide breadth of experts who focus on the various aspects of the TME with the intent of creating a more complete picture of the potential therapeutic landscape. The unique feature of this meeting is its breadth of experts focused on both the systemic and local TME, who also recognize varied paracrine interactions with heterogeneous neoplastic cells. Presentations will focus on emerging studies to enable attendees access to senior investigators, as well as new investigators spanning the diversity of disciplines called “cancer biology.” In addition to invited speakers, time will be allotted for short oral presentations selected from submitted abstracts and three integrated poster sessions.

Many oncogenes and tumor suppressor genes have been identified and mapped to signaling pathways that regulate cell growth and death. Dysregulation allows oncogenes and tumor suppressor genes to transform mammalian cells and cancers arise in conditions where multiple such events occur in the same cell. Many proto-oncogenes and tumor suppressors have been mapped to canonical signaling pathways, providing a more holistic view of growth control mechanisms and paving the way for systems level analysis of cancer mechanisms.It has also been found that a subset of the cells present in a tumor, called cancer stem cells, retain the ability to self-renew and to give rise to all cell types in a particular cancer.

An emerging concept is that tumors also strongly depend on external signals for maintenance and expansion. To fully understand tumor development and progression, a deeper knowledge of the cross-talk between tumor cells and their microenvironment and the interactions between cancer cells and cancer stem cells is needed.

Aims of event

This symposium brings together researchers from complementing fields to enhance our understanding of the communication between cancer cells and their microenvironment. Important open questions are: Which signals do cancer cells transmit to and receive from the stroma and how do these signals promote malignant growth? What roles does the extracellular matrix play during neoplastic transformation? What is the contribution of immune cells to tumor progression? How is the inflammatory response of the tumor microenvironment regulated by the microbiota?

Emerging research has provided evidence of a fundamental link between cytoskeletal structures such as centrioles and cilia in the regulation of DNA repair and genomic instability.

This is highlighted by an increasing number of cancers/syndromes with clear ciliary connections such as Birt-Hogg-Dube syndrome, Von Hippel-Lindau disease and Tuberous Sclerosis. Not only do cilia normally regulate many of the cellular signalling pathways commonly perturbed in cancer, but the ability to form cilia is lost in many transformed cells and it has been shown that the primary cilium can both mediate and suppress Hedgehog pathway-dependent tumourigenesis.

As the pivotal link between tumourigenesis and cilia function is emerging, this meeting will represent a timely opportunity to bring together researchers from the fields of cancer and cytoskeletal/cilia biology.

The format will allow invitation of speakers in cilia and cytoskeletal biology and speakers with specific interest in how these cellular processes influence the carcinogenesis process. Structured thematic sessions with talks describing fundamental cellular processes in cilia, cytoskeletal and tumour biology and how dysregulation of these processes can initiate disease, will culminate with talks aimed at the interplay between cilia, cytoskeleton and cancer and how these processes can be targeted therapeutically.
