Following the success of the 2016 conference in London, we present the Immuno-Oncology Summit Europe 2017. This is a unique inaugural conference dedicated to osteosarcoma in which researchers, clinicians and the patient community will interact. FACTOR stands for: Funding, Awareness, Collaboration, Trials, Osteosarcoma Research, which is the aim of the conference.It is planned to have inter-disciplinary discussions [involving pathologists, surgeons, geneticists, oncologists, radiologists] in three broad sessions : a session steered by clinicians on challenges in contemporary treatments, a session steered by researchers on innovative models and treatment approaches that are in progress or planned, with a final session steered by a mix of both, on the road ahead, and novel solutions to meet challenges and improve outcomes.The conference aims are to significantly improve osteosarcoma awareness and patient outcomes through collaborative disease management approaches, clinical trials and dedicated osteosarcoma research; to collectively brainstorm on the challenges and come up with collaborative solutions on a fast track; to create a symbiotic bridge between the osteosarcoma medical community and the patient community for an improved understanding of patient challenges and solutions; to provide a platform to various osteosarcoma advocacy/support groups to better collaborate and optimize their resources; to evaluate funding needs for research and provide impetus to fundraising strategies [by various advocacy groups] and be a dedicated forum to review progress on an annual basis on various osteosarcoma disease management initiatives.

Zusammenfassung
Defect in DNA damage repair and checkpoint control is the underlying mechanism for tumorigenesis, since it allows the accumulation of multiple genetic alternations, which are essential for the initiation of tumorigenesis. This has been clearly illustrated to be the cause of several human cancer-prone syndromes and also revealed by recent human genome studies. On the other hand, defective DNA repair and checkpoint activation also make cancer cells more vulnerable for particular DNA damaging agents or inhibitors that specifically disrupt some of these checkpoint pathways. With the increasing understanding of defects in DNA repair and checkpoint control in tumorigenesis, there are extensive interests in exploring these deficiencies, especially taking advantage of the synthetic lethality concept and targeting particular DNA repair and checkpoint pathways for cancer therapy. The purpose of this conference is to bring together basic, translational and clinical investigators and discuss the current and future directions, opportunities and obstacles in the development of these anti-cancer modalities and how to best apply these concepts in clinical practice.

Cellular responses to changes in oxygen availability have important implications for both physiology and pathophysiology, in particular in the setting of solid tumor progression. A number of hypoxia-inducible pathways have been characterized, such as the large transcriptome regulated by hypoxia-inducible factors (HIFs). However, many other oxygen-sensing pathways exist within the cell. The aim of this meeting is to disseminate up-to-date information on basic mechanisms of cellular and tissue hypoxia responses, and discuss the impact of these responses on physiology and cancer, with an emphasis on HIF-independent, but nevertheless fundamental, oxygen-sensing pathways. Outcomes of this meeting will include the dissemination of new scientific knowledge, the discussion of new methodologies, the education of graduate students and postdoctoral fellows in this discipline, and promotion of new collaborations between clinicians, basic researchers and industrial partners. This will be the eighth Keystone Symposia meeting on Hypoxia, which brings together academic, clinical and industrial researchers to enhance synergy between these groups and develop translational strategies for targeting hypoxic stress responses for therapeutic benefit.

In advanced prostate cancer, rapid and successful drug development has resulted in multiple treatment options, requiring careful decision making for individual patients. Clinicians face the increasingly difficult task of choosing from multiple potentially effective treatments that are also costly and potentially toxic. The inaugural Advanced Prostate Cancer Consensus Conference (APCCC) did provide a forum for discussion and debates on current treatment options for men with advanced prostate cancer. The conference concluded with voting by the panel on pre-defined consensus questions, targeting areas of primary clinical relevance. Following the conference a manuscript with containing the expert recommendations was published at Annals of Oncology.

Palliative radiotherapy is used to relieve our patients of pain or other distressing symptoms of their cancer and to improve their quality of life. It has traditionally been associated with the use of simple planning and delivery techniques but there is now increasing interest in the use of the latest treatment techniques such as Intensity-Modulated Radiation Therapy (IMRT) and Stereotactic ablative radiotherapy (SABR) to provide more effective and durable control and reduce toxicity. Alongside these developments, there has also been a change to a more multidisciplinary approach in the management of palliative cancer care with consultant therapeutic radiographers and nurses taking an increasingly active role in the planning of palliative radiotherapy.

While the importance of adoptive and innate immunity has been clear in the pathogenesis of human autoimmune disease resulting in a multitude of immune-based therapeutic approaches, the realization is now apparent that understanding immune evasion by cancer is central in developing curative treatments. This meeting will explore and contrast the underlying immune mechanisms resulting in autoimmunity and tumor evasion. The meeting is innovative in bringing together basic immunologists investigating mechanisms of tolerance with scientists exploring immune mechanisms of autoimmunity and cancer in both patients and experimental models. Thus, this Keystone Symposia meeting will cover the pathways in immunity and tolerance that lead to loss of immunological control, dysregulated immune responses and chronic inflammatory disease or tumor evasion. Presentations will include consideration of preclinical and clinical aspects of a diverse number of autoimmune and inflammatory diseases and cancer. Conference participants engaged in preclinical, translational and clinical research will hopefully engage in continuing conversations and collaborations which, over the long-term, will provide greater insights into the human immune response and allow us to reassess and further explore pathways that are driving autoimmune disease yet in opposition, lead to tumor evasion. Understanding checkpoints in autoimmunity and immune cell tolerance is important for delivering therapies to patients with autoimmune disease and cancer, and this meeting will provide a platform for the cross-pollination of clinical experience and experimental research. Attendees will have learned about the impact of targeted immune-based therapeutics on clinical outcome and, consequently, be able to widen their research scope accordingly.

Genome stability is the foundation upon which all cellular and organismal processes depend. This conference will grapple with the intricate array of biochemical reactions orchestrated by the cell to replicate, repair and segregate chromosomes accurately despite constant threats from spontaneous and environmentally-induced damage. Defects in these reactions lead to genomic instability, a confirmed driver of cancer and degenerative diseases. Fascinating and therapeutically crucial questions remain about the mechanisms underlying chromosome stability, and cross-disciplinary approaches are required to address them. How do epigenetic chromatin marks influence repair of underlying DNA sequences, and how can we predict the effects of chemotherapeutic agents that alter such marks? To what extent is the nucleus organized into subdomains with distinct functions; how are these subdomains altered when cells move or differentiate? How are the myriad helicases, nucleases and polymerases coordinated to safeguard genome stability, why are these molecules often mutated in human disease, and how can we ameliorate the effects of such mutations? What is the array of strategies available to cancer cells as they achieve unlimited proliferation? This conference has a history of bringing together investigators from diverse subfields who otherwise rarely meet. Cutting-edge concepts in translational, genomic, cellular, molecular, RNA and structural biology will be dissected to take the conversation to unprecedented levels of depth and breadth. The concurrent conference on “DNA Replication and Recombination” will provide additional opportunities for cross-talk. Both conferences are committed to nurturing interactions among longtime experts in the field with students, postdocs and investigators new to the field.

The central role of mitochondria in normal cell physiology is evident from human diseases, including metabolic disorders and aging, which are associated with changes in mitochondrial function. This SRC provides an interactive forum that covers the latest developments in the small GTPase field and presents the newest technological advancements that are being used to unravel the complex roles of small GTPases in human health and disease. Small GTPases (e.g. Ras, Rac, Rho, Cdc42, Rab, Rap, Rheb and Ral) are essential for key cellular processes such as cell differentiation, proliferation, and migration, as well as the regulation of subcellular events such as vesicle trafficking. These critical functions of small GTPases make them important participants in a wide variety of physiological and pathophysiological processes. The founding member of the small GTPase superfamily, Ras, is the most commonly mutated oncogene in human cancers, and many other small GTPases (including RhoA, Rac1, Rab5, Rheb, and RalA) also play fundamental roles in cancer development and progression. Due to these important oncogenic functions of small GTPases, there is a strong cancer theme underlying this meeting. However, small GTPases also participate in other biomedically important events, including the promotion of vascular diseases, immunological abnormalities, mental disorders, and metabolic conditions. For these reasons, progress from a number of fields in addition to cancer biology will be highlighted, including neurobiology, developmental biology, computational biology, and membrane biophysics to ensure that we are leveraging and learning from discoveries made in important related fields. The main objective of the meeting is to stimulate cross-pollination of ideas among researchers investigating basic biochemical mechanisms, cell and tissue biology, and translational models to define the diverse functions of small GTPases.

The structural and functional mechanisms of GRKs and arrestins, as well as their roles in biased/non-canonical signaling by GPCRs, in the actions of drugs of abuse, in cardiovascular disease and in cancer will be major highlights of the meeting. Sessions devoted to these topics will foster further studies to combat addiction and disease with the ultimate objective of positively influencing human health. The 9th European annual meeting of SERGS will be held in LILLE, France on June 14-16, 2017. The CRG (the French surgical group in gynaecological robot-assisted laparoscopy) is pleased to organise this famous meeting for the second time, the first time being in Marseille in 2012. Our annual French meeting will also be associated with SERGS in 2017. For years, teams have worked diligently on developing mini-invasive surgery, especially in gynaecology. Mini-invasive surgery, which includes conventional, single-port and robot-assisted laparoscopy, is the referent surgical approach (e.g., in endometrial cancer) because of a significant decrease in early postoperative morbidity. The rate of mini-invasive surgery for gynaecologic oncology in France in 2012 was 37%. The decrease in the curve of laparotomy in gynaecologic oncology began in 2007–2008.

2nd DNA Replication as a Source of DNA Damage Conference

The maintenance of genome integrity is critical for the suppression of cancer and premature ageing. Only recently has it become appreciated that DNA replication stress is a crucial driver of genomic instability. The timely progression of replisomes can be disrupted by lesions and secondary structures in the template, by bound proteins and by conflicts with the transcription machinery. A prolonged pause of the replisome then exposes single stranded DNA, which, due to its recombinogenic nature, can lead to genome rearrangements, fragile site expression and cell death. Importantly, some cancers present excessive endogenous levels of replication stress, which can be exploited for their clearance. In this conference, we aim to bring together scientists studying DNA replication and repair, with those interested in how DNA damage can influence cancer and ageing.

This SRC focuses on bringing together biochemists, geneticists, molecular biologists and clinicians interested in the pathophysiology of the leukemias and lymphomas, and the development of novel therapies for these diseases. Leukemia and lymphoma has provided a number of key paradigms for molecularly targeted cancer therapy, as illustrated by the remarkable success of all-trans-retinoic acid for the treatment of acute promyelocytic leukemia and tyrosine kinase inhibitor therapy for chronic myelogenous leukemia. New and promising small-molecule compounds and antibodies targeting epigenetic regulators, surface receptors and downstream signaling pathways are currently under development and will be discussed at this meeting. Added to these examples of targeted therapy, immunotherapy, including checkpoint inhibition and modified T cell-based therapies have emerged as important future anti-leukemia/lymphoma treatments.

The program will link detailed understanding of protein structure and biochemical mechanism to control of large-scale processes like the cell division cycle, cell morphogenesis, and metabolism. This SRC will explore the field’s latest research, which is revealing surprising mechanisms of regulation, previously unappreciated subcellular organization, logic of pathways that coordinate diverse intracellular systems. Importantly, derangement of kinase signaling underlies disease, including cancer and diabetes. Drugs targeting kinases and phosphorylation have achieved dramatic cures and promise more, and the conference has consistently offered a robust platform for clinically important work.

Over the past few years, the “epigenetic” regulation of the genome has become increasingly important to understanding both the etiology and fundamental mechanisms of aging and age-related diseases. Key to epigenetic regulation are two classes of lysine-modifying enzymes, the histone deacetylases (sirtuins and HDACs) and the histone acetyltransferases (HATs). Both classes of enzyme are critical for maintaining youthful gene expression patterns, cell cycle progression, DNA repair, stem cells, mitochondria, cell fate, differentiation, cognition, energy utilization, and the pathogenesis of most age-related diseases. Alterations in sirtuin and HDAC function have been linked to neurodegenerative disorders, muscle wasting, cardiac hypertrophy, cancer, HIV infection, and more generally the process of aging. Several HDAC inhibitors have been approved by FDA to treat various cancers. Small molecule modulators of sirtuins are currently in clinical trials for the treatment of cancers, neuromuscular disorders, inflammatory and metabolic disorders. With particular relevance to aging, the sirtuins, a subclass of HDAC, were first identified in yeast genetic studies as regulators of lifespan and the health benefits of dietary restriction. Activation or overexpression of sirtuins have been shown to protect mice and primates from diseases of aging, and extend mean and maximum lifespan of mice. Therefore, a thorough understanding of sirtuins and HDACs is required, not merely for understanding the regulation of chromatin structure, gene regulation and protein function, but also because these enzymes are intimately involved in normal and abnormal cellular processes that greatly impact human health. Although targeting HATs for therapeutic purposes has been relatively slow compared to targeting deacetylases, exciting new progress has also been made in recent years. Importantly, targeting the bromodomains present in many HATs has attracted a lot of attention lately.

EUS-ENDO will offer one day of live transmission from Paoli-Calmettes Institute to the Villa Méditerranée in the new center of Marseille. We have invited worldwide experts on EUS and Endoscopy, they will give lectures and will demonstrate the last developments of these technologies (ERCP, ESD, Therapeutic EUS, etc….) during the live demonstrations. New scopes will be showed as new accessories. The main goal of this meeting will be to demonstrate the complementarity of EUS and ERCP for bilio-pancreatic diseases, to show also new procedures as radiofrequency ablation of bilio-pancreatic cancers, cholangio-pancreatoscopy, confocal laser endomicrsocopy, etc…

The second day will be dedicated to hands on, 6 stations will be available for ERCP, EUS-FNA, T.EUS, GI bleeding management, EMR &ESD. This second day should be interactive between the participants and the experts, 4 workshops will be organized and also 3 sessions of oral communications in which participants will debate with the experts. Abstracts presented during EUS-ENDO will be published in Endoscopic Ultrasound review.

It will cover presentations from cancer genome projects, the areas of cancer functional genomics, systems biology, cancer mouse models and the translation and clinical impact of scientific results obtained. European Cancer Genomics research is particularly strong in the Heidelberg area, with three International Cancer Genome Consortium (www.icgc.org) projects being currently pursued with significant participation from Heidelberg institutions, including EMBL. This conference is a planned quadrennial event of these four associations and all other sub specialty associations to improve the quality of cancer care through multidisciplinary interactionand will be held in various parts of the Indian subcontinent once every four years. The ICC2017 conference is scheduled to be held from 9th to 12th November 2017 (with pre-conference workshop held on 8th Nov 2017) in Bengaluru, India.The theme for the conference is “INSIGHT, INNOVATION, INTEGRATION” The field of oncology is rapidly evolving and is in constant flux. What is thought to be true today may very soon be turned on its head by new insights and new innovations. It is therefore prudent and necessary that we the oncology community – periodically stop in our fast tracks and take stock of some important insights and innovations and debate how best we can integrate these innovations into our practice.
