 Zusammenfassung Cellular functions depend on numerous protein-coding and noncoding RNAs and the RNA-binding proteins (RBPs) associated with them, which form ribonucleoprotein complexes (RNPs). Mutations that disrupt either the RNA or protein components of RNPs or the factors required for their assembly can be deleterious. The discovery of disease-causing mutations in RNAs or their interacting RBPs not only adds to our growing understanding of RNA biology and chemistry, but also offers new therapeutic targets and contributes to ongoing efforts in developing mRNA-targeting antisense and RNAi therapeutics. This meeting focuses on a general understanding of post-transcriptional gene regulation and how perturbation of complex networks of RNA-protein and RNA-RNP interactions triggers disease.

 Zusammenfassung The lymphatic vascular system is essential for lipid absorption, fluid homeostasis, and immune surveillance. Malfunctions of the lymphatic system can lead to congenital or inherited disorders such as lymphedema, a disfiguring and disabling disorder that is caused by imbalance in lymph absorption and often characterized by swelling of the extremities.

Our understanding of the genes and mechanisms controlling the formation of the lymphatic vasculature has improved a great deal during the last decade. The availability of molecular markers and animal models with various degrees of lymphatic defects has been instrumental to the progress in this field. Also, work in the last few years had led to a better understanding of the functional roles of this vascular system in health and disease. Furthermore, recent work has also unraveled additional functional roles of the lymphatic vasculature in fat metabolism, obesity, inflammation, tumor lymphangiogenesis, and the regulation of salt storage in hypertension.

The 2012 Gordon Conference on Molecular Mechanisms in Lymphatic Function & Disease will bring together an outstanding group of basic and clinically-oriented interdisciplinary scientists to discuss their latest progress in a wide range of topics in this rapidly advancing field of research. The program contains sessions about lymphatic vascular development, lymphatics and inflammation, lymphangiogenesis and cancer, lymphatic-related diseases such as obesity, hypertension, immune suppression, congenital and secondary lymphedema, and therapeutic approaches. The goal is to discuss new insights into the cellular and molecular mechanisms mediating lymphatic development and function, and the design of new interventions against pathological conditions affecting the lymphatic vascular system.

The Conference will bring together investigators who are at the forefront of their field, and will provide opportunities for junior scientists and graduate students to present their work in poster format and exchange ideas with leaders in the field. Some poster presenters will be selected for short talks. This conference is intended to provide a unique format for the exchange of ideas and information, to discuss the latest research findings and technical advances in a collegial atmosphere.

 Zusammenfassung The 2012 Gordon Conference on Protein Transport across Cell Membranes is organized around a theme of merging traditional approaches of cell biology, biochemistry, and biophysics, with emerging technologies of structural biology and single molecule fluorescence imaging to address the same or similar fundamental questions. Topics to be covered include protein transport and membrane protein integration into the ER by the Sec machinery and also by Sec-independent mechanisms, multiple pathways for protein export and secretion across prokaryotic membranes, import and routing of proteins into chloroplasts, mitochondria, and peroxisomes by both novel and evolutionarily conserved mechanisms. Sessions will also cover non-traditional protein translocation mechanisms. This Gordon Research Conference will foster the synthesis of new ideas by bringing together leading international speakers with investigators at all stages of their careers. In addition to invited speakers, short talks will be selected from submitted abstracts.

The 2012 conference also features a pre-conference Gordon Research Seminar (GRS), organized by postdoctoral scientists, that will be oriented towards junior investigators to provide them with background on common mechanisms involved in protein transport, to discuss their ongoing research projects, and to facilitate interaction and networking with their peers within this scientific community. We fully anticipate that the talks, discussions, poster sessions, and other informal interactions at this GRC and GRS will contribute to advancing our understanding of molecular mechanisms of protein trafficking, which is essential to both prokaryotic and eukaryotic cells.

 Zusammenfassung During the past 10 to 15 years, there has been significant progress in our understanding of pancreatic islet development and islet biology. This knowledge has been complemented and accelerated by major advances in diverse fields including islet transplantation, stem cells, small molecule screening, high-throughput sequencing, epigenetics and human genetics. The integration of these divergent disciplines has allowed us to develop a clearer definition of a functional b cell and has elucidated many of the mechanisms underlying the differentiation, survival and death of the b cell. Furthermore, these studies have revealed the potential to generate new b cells through replication or neogenesis. Improvements in differentiation and transdifferentiation protocols have also illustrated the potential of using stem cells and some somatic cell types as sources of islet cells, but these studies have also revealed the challenges associated with generating functional b cells. The purpose of this Keystone Symposia meeting is to highlight the areas of islet biology that have seen the most remarkable advances and to address some of the controversies that still exist in the field, especially with regards to the generation and maintenance of functionally mature b cells for therapeutic purposes. In addition, where it is possible, comparative mouse and human studies will be discussed to illustrate if and when mouse studies can be applied to the human situation.

 Zusammenfassung Cells are continuously subjected to a barrage of DNA damage from both endogenous and external sources, and an elegant set of enzymatic pathways has evolved to repair or tolerate this damage. Deficient or aberrant repair results in mutations that can lead to cancer.

The 2012 conference on DNA Damage, Mutation, & Cancer is designed to address diverse research areas focused on DNA damage, mechanisms of mutagenesis and genetic instability, and the relationships between genetic instability and human cancer. Cutting edge research will be presented on the molecular mechanisms involved in the repair and processing of DNA lesions, genome instability, and cancer etiology. Topics to be covered include DNA structure and mutagenic mechanisms, telomere biology and genetic instability, DNA repair deficiency syndromes and cancer, pathways for damage repair or tolerance through lesion bypass, chromatin-mediated genomic instability, and promising strategies for targeting DNA repair pathways in cancer therapy.

The invited speakers, who are at the forefront of their respective fields, come from a variety of disciplines, and the program will be designed to allow ample time for discussion following each presentation, as well as during the poster sessions. The conference will strongly encourage the presentation and discussion of unpublished results. We will also provide an opportunity for junior investigators to present short talks on exciting topics to be selected from submitted abstracts. We anticipate that productive scientific exchange at this conference will significantly impact cancer research and result in establishing novel multi-disciplinary collaborative efforts.

 Zusammenfassung It has become apparent that RNA plays a major, but still largely unexplored, role in the regulation of gene expression. It is becoming increasingly evident that longer forms of non-protein-coding RNAs (ncRNAs) are involved directly in regulating gene expression and epigenetic states. These long ncRNAs appear to be functioning as an endogenous RNA regulatory system, playing a major role in the epigenetic control of differentiation and development. These observations, across various model systems, strongly suggest that long ncRNAs are more intricately involved in the regulation of gene expression than previously envisioned. The goal of the Keystone Symposia meeting on Non-Coding RNAs is to highlight the similarities and differences in various model systems to better understand the role of long ncRNAs in the regulation of genomes. This is an entirely new emerging field of study that is on the forefront of modern biology. Opportunities for interdisciplinary interactions will be significantly enhanced by the concurrent meeting on Eukaryotic Transcription, which will share a plenary session with this meeting.

 Zusammenfassung Gene expression in eukaryotes is regulated primarily at the level of transcription. Biochemical, genetic and molecular genetic analyses of a handful of genes have provided a basic understanding of transcription. Advances in proteomics have rapidly expanded the repertoire of proteins and protein-protein interactions that participate in gene expression, and genome-wide analyses of protein distributions across genomes and gene expression are expanding and challenging our knowledge. For example, RNA polymerase II is found concentrated at the promoters of thousands of genes, causing much interest in the field to shift focus to the role of elongation in regulating gene expression. The functions of vast noncoding, transcribed regions of the genome are largely unknown. HDACs, thought to function primarily as repressors, tend to be associated with highly transcribed genes. This meeting will explore new and exciting aspects of eukaryotic gene regulation emerging from genomic, proteomic and other high-throughput techniques.

 Zusammenfassung Nuclear receptors (NRs) represent a large superfamily of transcription factors that function as ligand-dependent sensors for a diverse set of fat-soluble hormones, vitamins and dietary lipids. Together, NRs govern expression of genes involved in a broad range of reproductive, developmental, metabolic and immune response programs. For this reason, NRs have been investigated for over 20 years as promising targets for drugs to treat a multitude of therapeutic indices, including cancer, diabetes, obesity, cardiovascular disease, hypertension, inflammation and cholestasis. However, despite the success of numerous NR-based therapies, there remain significant hurdles exploiting NRs to develop newer classes of therapies that achieve targeted benefits with reduced side effects. Thus, understanding the mechanisms, transcriptional targets and networks of physiologic pathways that govern NR action has become an important new strategy for evaluating the role of NRs in health and disease. This ?reloaded? effort is now beginning to produce exciting new avenues of research in the field. This meeting will bring together a key group of junior and senior investigators that are exploring these new directions and provide participants with a matrix of unique possibilities for them to investigate in both academic and pharmaceutical laboratory settings.

 Zusammenfassung The 2012 Myelin Gordon Conference will bring together researchers in PNS and CNS myelin biology to present the latest advances and discuss key questions in the field. Tentative session topics include the development of myelinating glia, the extrinsic signals and signaling pathways that positively and negatively regulate myelination, regulation of axon function, integrity, and organization by myelinating glia, and investigations into myelination and remyelination in the adult, including the role of stem cells. Clinical relevance will be stressed throughout the conference, focusing on various myelin disorders and strategies for myelin repair. Invited speakers will be drawn from a variety of scientific disciplines, including developmental, cellular and molecular biology. Senior investigators will serve as session chairs to guide the discussion and provide overviews of specific topics, in addition to presenting new findings. There will be a concerted effort to invite investigators at the junior faculty and postdoctoral levels to speak at the meeting to enhance their visibility and foster scientific interactions. Members of under-represented minority groups are especially encouraged to attend. This year, the conference will be held in conjunction with the Myelin Gordon Research Seminar, which will focus on myelination and remyelination in the CNS .  Zusammenfassung The Fibroblast growth factors (FGFs) comprise a large family of growth and differentiation factors with essential functions in development, metabolism, and repair of various tissues and organs. Dysregulation of FGF signaling is associated with a wide variety of human diseases, including several developmental/genetic diseases, metabolic disorders, and malignancies. This conference will encompass the most recent and timely research in this highly diverse field, and promote interdisciplinary interactions between scientists, clinicians and colleagues from industry studying all aspects of FGF biology. Topics will cover Biochemistry of the FGF signaling complex and control of downstream signaling pathways and their impact on cell biology, physiology, endocrinology, development, pharmacology, pathology and regeneration/repair.

In addition to presentations by invited speakers, abstracts will be selected for shorter oral presentations for each session, ensuring that the program includes a breadth of exciting new topics emerging in the period just preceding the conference.

Strong emphasis will be placed on presentation of unpublished data and extended discussion times tackling the cutting edge of the field. Poster sessions will also be given high priority. Applications will be accepted from a diverse mix of established investigators, junior PIs, postdocs and graduate students. There will also be a satellite GRS meeting for postdocs and graduate students prior to the main GRC.

 Zusammenfassung The cyclic-nucleotides cAMP and cGMP are intracellular chemical signals that control most aspects of cellular behavior. These so-called "second messenger" responsive pathways have long been considered therapeutic targets for the treatment of cardiovascular and inflammatory airway diseases and neuropsychiatric disorders. The 2012 Gordon Conference on Cyclic Nucleotide Phosphodiesterases will convene in Il Ciocco Italy from May 20-25, 2012. This 8th meeting in the series will provide a forum for investigators from academia and industry to present cutting-edge research and ponder the latest concepts in molecular and cellular cyclic nucleotide metabolism and cell signaling. The opening session of the meeting will set the scene with lectures from three renowned experts in cyclic nucleotide signaling. Pharmacological intervention of second messenger responses as therapies for a variety of pathophysiological disorders will be a featured topic of discussion. The scientific program will also feature a range of lectures that explore the structural biology of phosphodiesterases, the spatial organization of cAMP/cGMP effector molecules, genetic manipulation of phosphodiesterases and the pathophysiolgical implications of defective cyclic nucleotide signaling. The keynote lecture will be delivered by Dr. Johannes L. Bos on the interface of cAMP and cancer signaling pathways. The organizers encourage junior scientists and graduate students to attend. Delegates should submit an abstract of their research for poster presentation and selected individuals will be invited to give short talks on emerging aspects of cyclic nucleotide research. The collegial atmosphere of this conference, with discussion sessions and informal gatherings in the afternoons and evenings, creates an optimal environment for scientists from different disciplines to interact.

 Zusammenfassung Chemotactic Cytokines 2012 will be our 10th GRC. This is a good juncture to look back and commemorate a series of unforgettable meetings, all marking significant milestones of the extraordinary journey in time by the molecules of our dreams, chemokines. From their humble beginnings on the fringes of experimental research as a couple of leukocyte chemoattractants, chemokines became one of the largest families of coherently functioning, versatile intercellular signals, studied now by the cutting edge mainstream molecular science and medicine. It is currently clear that chemokine activities contribute to almost every aspect of our biological existence, starting with the moment of our conception, through months of intrauterine confinement; protecting us during the years of growth, health and wellbeing, only to push us later on, along the countless avenues of diseases, ultimately into the grave. As much as it is almost impossible to encompass the breadths of all chemokines’ activities, it became also impossible to cover all the aspects of contemporary chemokine research, even at a comprehensive conference, such as GRC. Instead of attempting this, Chemokine GRC 2012 for the first time will have a specific thematic focus, also reflected in its title, Chemotactic Cytokines: Molecular and Cellular Mechanisms in Health and Disease. This means that, rather than restricting in any way the breadth of chemokine implications covered, our GRC will concentrate primarily on their most fundamental, mechanistic aspects.

The research on chemokines has considerably accelerated and spread from initially few principal centers to thousands of institutions worldwide. Hence, nowadays it is hard to predict ahead of time in which laboratories the hottest and the coolest chemokine discoveries of 2012 will emerge. In anticipation of such discoveries and to conform with the spirit of the Gordon Conference, calling to showcase cutting edge unpublished data, we allocated even more speaker slots to the late breaking and short talks selected from abstracts. These should complement invited lectures by top scientists covering biochemistry of chemokines and the cellular responses to them, in vitro and in vivo mechanisms of chemokine induced migration, chemokines in immune organogenesis and immune organ function, chemokines in infection and immunity and the contribution of chemokines to the disease pathogenesis, including cancer.

During the last several years a new generation of young scientists joined the research on chemokines. Students and post-docs constitute now the most numerous group of Chemokine GRC participants. The traditional GRC format provided for their top notch and broad ex cathedra education, but not necessarily fostered their most active participation. This hiatus was addressed and, with considerable pride and excitement, we are announcing that Chemokine GRC 2012 will be twinned for the first time with the Chemokine Gordon Research Seminar, a satellite meeting for young bench scientists, post docs and students, which will take place May 26-27, 2012. This meeting will allow young chemokine researches to interact in an informal setting, exchange ideas, gain confidence as speakers and achieve higher personal visibility. Moreover, it will provide a forum for their concerted mentoring by the most outstanding chemokine scientists.

Our GRC will be returning to Il Ciocco, resort tucked away amidst Toscana Mountains, in the heart of a fascinating land, rich in history and culture, warm, hospitable and welcoming. Despite Il Ciocco being not only one of the most beautiful, but also largest GRC venues, its capacity is finite and only first 200 registered participants will be able to attend the Chemokine GRC, 2012. Our last GRC was oversubscribed with long waiting lists and considerable disappointment for those at end of the queue who could not attend. To assure your participation please register early!

 Zusammenfassung The 2012 Gordon Conference on Muscle: Excitation/Contraction Coupling will present results of the latest research on the intricate processes that link excitation to contraction in skeletal and cardiac muscle as well as the disorders that result from their malfunction. The conference will emphasize the many multi-disciplinary approaches used to study this process, including structural biology, electron microscopy, biochemistry, biophysics, molecular genetics, cell physiology, and animal studies.

Invited speakers will discuss and evaluate critically new findings on (1) the structure of the molecular machinery underlying excitation-contraction coupling, (2) the rapid calcium signaling important for excitation-contraction coupling as well as the slower calcium signals that are important for regulating other processes in striated muscle, (3) the effects of human disease-causing mutations, fatigue and aging on excitation-contraction coupling (4) the roles of both the primary players and the accessory proteins of the triad junction in excitation-contraction coupling, (5) the role of calcium signaling in adaptation, and disease progression, and (6) the development and implementation of new methods to probe muscle excitation-contraction coupling and signaling.

In addition to the invited speakers, the organizing committee will select several poster presenters to give short talks during the formal sessions. The conference will stimulate discussions and will provide a forum that will give young and established investigators the opportunity to meet and interact and exchange their opinions on various topics of the meeting. By bridging basic science, translational and clinical studies related to the mechanism of muscle diseases, this meeting will foster new research directions and scientific collaborations moving the field forward into the future.

 Zusammenfassung The Gordon Research Conference on NOX Family NADPH Oxidases meets biannually, alternating between US and European sites so as to attract a broad range of international scientists active in this rapidly growing field of biomedical research. The 2012 meeting will also include the first NOX Gordon Research Seminar for research trainees. The NOX GRC will feature cutting-edge unpublished work presented by experts across the NOX field, whereas the NOX GRS will be geared specifically to the interests and needs of young research trainees in the field, primarily graduate students and post-doctoral fellows.

NADPH oxidases of the NOX family have in recent years been shown to comprise the predominant source of reactive oxygen species (ROS) produced by most types of cells. It is abundantly clear that the science in this field is moving very rapidly, with many exciting advances in understanding of the basic biochemistry and cell biology of the NOX family of enzymes, particularly the contrasting properties and functional roles of the seven discrete isoforms comprising the family. Moreover, NOX-generated ROS are directly involved in a wide range of both physiologic and pathologic processes. Many linkages between NOX enzymes and disease pathogenesis are being elucidated and this knowledge is dramatically accelerating advances in the identification of new and often unexpected therapeutic targets and development of disease-modifying pharmacologic agents. The NOX Gordon Conference program has been clearly established as the top forum for the presentation and discussion of the latest advances in this field. Thus, the 2012 NOX GRC/GRS meetings promise to be timely, exciting, and paradigm-challenging.

The program for the meeting has been developed around the theme of "NOX Biology and its Translation to Human Disease and Therapy." The preliminary conference program comprises nine scientific sessions, four with basic themes and five with a disease-oriented translational emphasis, including one session devoted entirely to recent exciting advances in the development and testing of small-molecule NOX inhibitors. Each session features a discussion leader and three to four speakers, plus a few young investigator presentations to be selected from among an anticipated ~100 poster abstracts. There will be ample time for vigorous discussion after each talk. The GRS program will feature speakers selected from among trainee abstracts, as well as career development components and mentoring by senior scientists. Attracting a diverse group of participants is a major priority, as reflected for example in the high prevalence of women scientists (35%) listed in the preliminary GRC program.

Les Diablerets provides an intimate setting in the Swiss mountains for the 2012 GRC on Immunochemistry and Immunobiology, where a maximum of 150 participants will exchange ideas and discuss the latest advances in the field. In contrast to many conferences that focus on specialized Immunology topics, this 2012 GRC on Immunochemistry and Immunobiology will feature broad coverage of both innate and adaptive immunity, and will span the full spectrum of basic mechanisms to applied research. The meeting will be divided into nine half-day sessions, with approximately 36 invited internationally recognized speakers.  Zusammenfassung First organized in 1994 to focus on nitrogenase and its unique iron sulfur chemistry, knowledge about iron sulfur clusters has grown exponentially in the last two decades, and in 2006, the name of the conference was accordingly changed to Iron Sulfur Enzymes. The conference has tremendous breadth, featuring sessions on numerous iron sulfur enzymes of unicellular organisms, animals, and plants, and important insights into how these versatile protein cofactors are synthesized and function. Proteins containing FeS cofactors perform a variety of biological functions ranging across electron transfer, acid-base catalysis, and sensing where they are agents for cell regulation through transcription (DNA) or translation (RNA). They are redox catalysts for radical-based reactions and the activation of H2, N2 and CO2, processes that offer scientific and economic challenges for industry. Whereas initial meetings concentrated on the roles of cyclic nucleotides as second messengers, the conference has expanded to include many mechanisms by which cells respond to extracellular stimuli, with emphasis on protein kinase and heterotrimeric G protein-dependent networks directly regulated by cell-surface receptors. Malfunctions in these signaling pathways contribute to health problems that affect millions worldwide. Consequently, cell-surface receptors represent major pharmaceutical targets, and a greater understanding of the signaling networks they control will provide many future opportunities for novel and more selective therapeutic intervention.

To this end, the 2012 meeting will bring together world leaders and early stage investigators to facilitate the exchange of ideas and accelerate the pace of research in the signal transduction field. Sessions will cover recent advances in the study of heterotrimeric G protein and protein kinase signaling pathways, how these pathways contribute to disease, and how they might be targeted through translational research. During the last 20 years, work in many laboratories has led to the identification and understanding of many of the key players involved in apoptosis.  Zusammenfassung Research on C.elegans plays an influential role across biology and has been at the forefront of genetics. Genetic analyses in C.elegans have led to advances in understanding many processes conserved throughout the animal kingdom, including establishment of cell polarity, developmental patterning, cell death, RNA interference, ageing, axon pathfinding, neuronal signaling and many others. Evidence of the impact of C.elegans research is the award of three Nobel prizes and a Lasker Award.

Two types of meetings focus on C.elegans research. The International C.elegans meeting, organised once every two years in the USA, covers all aspects of C.elegans research. The large size (over 2000 attendees) and diversity of the International meeting does not allow easy discussion or interactions among groups who share interest in particular research areas. To fill this gap, C.elegans “Topic meetings” are held in alternate years with the International meetings. These are focused meetings on particular aspects of C.elegans biology. Topic meetings provide a vibrant and exciting arena for discussion of current research, and the sharing of new ideas, methods, and reagents.

It is the team work of proteolytic enzymes and their inhibitors which makes a go of many cellular functions. We recognize proteolytic enzymes as managers that catalyze the most important and ubiquitous post-translational modifications in cells. Irreversible and uncompromising, proteases regulate life and death of every organism. Diseases caused by mutations in the genes of proteases or their inhibitors, threats introduced by bacterial, fungal or viral proteases taught us to look more closely on the hundreds of diverse proteases and the many inhibitors interacting with them to direct cellular functions.

Evolution was clever enough to come up with an excellent system of cellular decision makers, but the system can be cheated in the most mischievous way arthritis, cancer, diabetes, obesity, malaria, neurodegenerative disorders and many more can be caused by stray proteolytic enzymes and their inhibitors. Understanding the structures and functions of proteases, watching proteases in action, and approaching their intricacies in the protease web is a must but more so, it is a hope for designing new drugs and better therapeutic treatments based on translational research. On the bright side of life, proteases enable development and maintain physiology. Proteases and their inhibitors are clearly sustainable, they are vital.

 Zusammenfassung The Gordon Research Conference on Lysosomes and Endocytosis covers recent advances in understanding the function and molecular regulation of the endocytic pathway, lysosomes, and related subcellular organelles. Endosomes and lysosomes play critical roles in the physiology of cells and organisms by serving as the cellular entry point for nutrients, toxins and pathogens and the nexus for intercellular signaling networks. Many inherited and somatic diseases result from defects in endocytic and lysosomal pathways. Some pathogens are destroyed by these pathways, while others subvert their function during infection. Moreover, these pathways play key roles in the stimulation and implementation of immune responses, glucose and lipid metabolism, cell and tissue development, and cancer.

Topics of focus for the 2012 meeting include molecular mechanisms and cell biology of endocytic and degradative pathways, as well as their role in human health and disease. Sessions will cover molecular features of endocytic membrane traffic such as coat formation, membrane tubulation, and cell physiology including endosomal and lysosomal membrane traffic, autophagy and formation and function of specialized lysosome-related organelles. Other sessions will focus on the roles of endocytic pathways in infection and immunity, diabetes and metabolism and development. The conference will continue to feature a session on 'New Directions' in which novel technical and conceptual developments in the field will be discussed.

The meeting will be divided into nine half-day sessions, comprising a mixture of invited speakers and those selected from abstracts. Attendance by under-represented minorities, students, postdoctoral fellows, junior faculty, and faculty from primarily undergraduate institutions is encouraged. If you are a student or postdoctoral researcher, it is very important to identify the principal investigator of your laboratory in your application. This conference is usually oversubscribed, so early application is advised with a suggested deadline of April 1, 2010, after which the conference may be filled. Applications received after the conference is full will be assigned to a waiting list.

Prior to and in association with the Gordon Research Conference, new in 2012 there will be a Gordon Research Seminar on Lysosomes and Endocytosis, in which students and post-doctoral researchers can present their work to each other and attend instructional seminars prior to the arrival of senior investigators. The scientific program being developed will include topics such as Predicting drug metabolism using P450 structural information. Other topics include the importance of Environmental factors, such as seasonal variation and our gut genome, on drug metabolism. Time dependent inhibition of drug metabolism will also be covered. New tools for ADME studies such as stem cell derived tissue-specific cells and various Humanized mouse models will also be discussed. Other topic areas include metabolomics linking transporters and drug metabolizing enzymes to new metabolic pathways. A final focus will be new frontiers, new techniques, methodologies, and data analysis. The program will continue to follow the traditional GRC Drug Metabolism format with a Keynote lecture Sunday evening, four morning sessions - each with four speakers, and three evening sessions with three speakers each. The Wednesday evening program will feature a mix of six graduate students and post-doctoral fellows who will deliver oral presentations that are selected from the many outstanding posters submitted. Vice Chair James Mangold will organize the poster sessions and attendees are encouraged to submit posters. As always, this meeting offers a wonderful opportunity to learn and discuss unpublished research, discuss new ideas, and establish collaborations with senior and junior scientists from academia and industry. Of course we will maintain the traditions passed down from generations of Drug Metabolism Gordon Conferences including the historic Tuesday afternoon Academia vs. Industry softball game (bring those gloves!), Monday afternoon tennis and Wednesday afternoon golfing, and there are always hiking and canoeing outings in the beautiful White Mountains of New Hampshire.

All participants are asked to bring a poster since approximately ten abstracts will be chosen for short talks. We strongly encourage graduate students and post doctoral associates to apply to both the 2012 Ion channels GRC and GRS. In addition, selected abstracts from these "early-career" investigators will be chosen for a "Data Blitz", in which the presenters will have two minutes and a slide to present their key findings that can be further discussed during the GRC poster sessions.

A main goal of the meeting is to stimulate detailed discussions among all attendees, giving rise to new ideas and directions for the field. Interactions are promoted by programming a generous amount of time for discussion sessions and by providing opportunities for informal gatherings at meals and in the afternoons and evenings. The 2012 meeting will be held at Mt. Holyoke College in idyllic Western Massachusetts. Mt. Holyoke College is within easy driving distance from Boston's Logan Airport, and a shuttle bus will provide transportation prior to and at the conclusion of the meeting. This GRC provides an outstanding forum to present and discuss new findings about the mechanisms and regulation of RNA metabolism in normal and diseased cells and tissues. The emphasis will be on the post-transcriptional transactions during the elongation, splicing, polyadenylation, transport, translation, and decay of mRNA as influenced by regulatory proteins and non-coding RNAs including microRNAs. A broad cross section of methodological approaches including genomic and bioinformatic strategies, single molecule and single cell-based studies, in situ localization, including genetic approaches with model systems as well as molecular phenotypes associated with human disease states. transcriptional coupling, are applied and discussed. Elucidating the basic mechanisms of post-transcriptional gene regulation is essential to gain a full understanding of the organization, function, and evolution of the human genome, and other genomes, as well as of the extensive involvement of RNA-processing dysfunction in numerous genetic and acquired disorders.

The meeting will convene approximately 45 speakers representing key areas of post-transcriptional gene regulation, with a total of 150-180 participants. The program will consist of nine morning or evening sessions that will broadly address cutting-edge issues in the following areas: Nuclear RNA Metabolism: The Transcription Connection; Bioinformatics: Gene Expression and RNAomics, including the evolution of RNA-processing signals and factors; RNA Splicing Catalysis, Regulation and Fidelity; Non-coding RNAs; RNA Transport and Localization; RNA Turnover, including nonsense-mediated mRNA decay and other types of quality control; Translational Control; RNA and RNP Structure; and RNA-processing Dysfunctions in Disease. This emerging field of science attracts a young cohort of scientists who get a chance to interact closely with established leaders, promoting a creative environment of inquiry that greatly advances the field.

Early efforts of the single molecule community were almost entirely focused on developing new techniques. Now, and in large part due to this conference, single molecule techniques are rapidly occupying a central role in biological research at all levels. This transition was made possible by the availability and dissemination of robust techniques that use fluorescence and force probes to track the conformation of molecules one at a time, in vitro as well as in live cells. Single-molecule approaches have changed the way many biological problems are studied. These novel techniques provide previously unobtainable data on fundamental biochemical processes that are essential for all forms of life. The ability of single-molecule approaches to avoid ensemble averaging and to capture transient intermediates and heterogeneous behavior renders them particularly powerful in elucidating mechanisms of the molecular systems that underpin the functioning of living cells.  Zusammenfassung The synapse serves as the basic signaling unit of the nervous system. Information transfer at the synapse, called synaptic transmission underlies every aspect of brain function and is relevant to most neurological diseases, as well as mental illness and drug addiction. The communication between neurons is a highly complex, dynamically regulated process and a comprehensive understanding of such a process requires a multidisciplinary approach and the complementary use of cutting-edge techniques. The Gordon Research Conference on Synaptic Transmission, established in 1998, highly successful and widely attended since then, has developed into the quintessential biannual meeting covering all aspects of synaptic communication.

The 2012 Conference will present major recent advances on a wide range of topics related to synaptic transmission including vesicle fusion, structure function relationships, novel forms of plasticity and integration of synaptic function into neuronal networks. The Conference will bring together a diverse group of investigators who are at the forefront of their field, and will provide ample opportunities for junior scientists and graduate students to present their work in poster format and exchange ideas with world-class leaders in the field. Some poster presenters will be selected for short talks. In addition, keynote lectures will be delivered by two outstanding scientists whose work on synapses and networks is both highly influential and complementary.  Zusammenfassung Many environmental toxins provoke a rise in cellular reactive oxygen species (ROS) that can damage cellular macromolecules, potentially triggering apoptotic signaling. However, ROS are also necessary components of growth factor, cytokine and immune signaling and may be involved in aberrant proliferative signaling that is the hallmark of cancer. Thiol-containing proteins and small molecules are important targets of ROS action, where reversible oxidation can act as a molecular switch to modulate function and regulate cell signaling pathways. Thiol oxidation is also crucial for protein folding, protein secretion and protein trafficking. While redox "imbalance" is widely regarded as the cause of ROS-dependent pathology, we have only a limited understanding of how ROS generation is associated with normal cell signaling. Furthermore, antioxidants have conflicting effects when used as dietary supplements or chemotherapeutics. The need has never been greater for research scientists, physician scientists, scientists from governmental agencies as well as those from industry to come together in this highly interdisciplinary setting to exchange ideas and perspectives and foster new collaborative research interactions. By bringing together investigators with varied expertise in biophysical methods, bioinformatics and animal model systems, with physicians focused on disease processes, the meeting is expected to further stimulate collaborations and catalyze scientific progress as has been exemplified by the successes of the previous meetings.

This conference is in its fourth cycle after three very successful meetings in the U.S. and Italy in 2006, 2008 and 2010. The highly international flavor of this meeting is reflected in the U.S.- and France-based chair and vice chair, respectively, and a diverse range of speakers from around the globe. Topics will include disulfide bond formation in intraorganellar protein folding, chemical biology of reactive oxygen species (ROS) generation and action, and how perturbations in the localized reactive oxygen, nitrogen and sulfur redox environment can alter signaling and lead to disease. Significant emphasis will be placed on cellular signaling dysfunction from environmental stresses and their link to cancer development. Participation by junior researchers in this conference will also be enhanced through a preceding Gordon Research Seminar with keynote speaker Mike Marletta and speakers and discussion leaders chosen from among the graduate student and postdoctoral community in this field.

 Zusammenfassung One-carbon (C1) compounds play a central role in microbial metabolism. C1 compounds include methane, carbon monoxide, carbon dioxide, and methanol as well as coenzyme-bound one-carbon compounds. They are of broad global importance because several of these (e.g., methane) are important energy sources, some (e.g., carbon dioxide and methane) are potent greenhouse gases, and others (e.g., dichloromethane) are xenobiotics. C1 compounds are central in pathways of energy metabolism and carbon fixation by microorganisms and many are of industrial interest. The 2012 Gordon Conference will present and discuss cutting-edge research in the field of microbial metabolism of C1 compounds. The conference will feature the roles and application of C1 metabolism in natural and synthetic systems at scales from molecules to ecosystems. The conference will stress molecular aspects of the unique metabolism exhibited by autotrophic bacteria, methanogens, methylotrophs, aerobic and anaerobic methanotrophs, and acetogens. Complementary perspectives will be provided by leading scientists in biochemistry, general microbiology, molecular biology, ecology, systems- and synthetic biology.

The Conference will bring together investigators who are at the forefront of their field, and will provide opportunities for junior scientists and graduate students to present their work in poster format and exchange ideas with leaders in the field.
