| Résumé | In the last decade, cytokine therapies have been included as the standard of care for the treatment of infectious disease and cancer, and cytokine antagonists have been deployed for the management of chronic inflammation. Many researchers in this field are interested in basic principles through which cytokines mediate communication with and between cells of the immune system. Others are interested in the contributions of cytokines and cytokine signaling to the development of, or protection from, a broad range of diseases including allergy, infectious diseases, cancer, atherosclerosis and autoimmunity. With the recognition that many of these pathways impact broadly on multiple conditions, the goal of the Keystone Symposia meeting on The Biology of Cytokines is to promote interaction between scientists who study the molecular mechanisms of cytokine function, signal transduction and gene expression, and scientists who study how cytokines work in vivo. We anticipate that this meeting will foster discussion aimed at translating this knowledge into novel therapies for human disease. Opportunities for interdisciplinary interaction will be significantly enhanced by the concurrent meeting on Th17 Cells in Health and Disease, which will share a keynote address and two plenary sessions with this meeting. |

Th17 cells have unique regulation and have been shown to be associated with many types of immune diseases and cancer. Since the first Keystone Symposia meeting on Th17 cells in 2009, there has been rapid progress in our understanding of the roles of IL-17-producing T cells in innate immunity to infection, the plasticity of Th17 cell, and genetic diseases associated with Th17 defects. The goal of this second Keystone Symposia meeting on Th17 cells is to gather experts in basic and clinical research areas and provide a productively interactive forum to further our understanding of Th17 regulation and function in immune responses and diseases. | Résumé | The impact of small non-coding RNAs has profoundly touched the fields of development and cell biology, functional genomics, human disease and drug therapy. This mode of gene regulation is not restricted to eukaryotes; bacteria utilize small RNAs, notably those made from CRISPR loci that silence the expression of bacteriophages, transposons and plasmids. There are still many gaps in our knowledge of the mechanisms used by small RNAs, particularly newly discovered RNAs. It has become clear that this field of study needs an annual forum to enable rapid dissemination of new discoveries between researchers. Overall, the goals of this proposed meeting are: (1) to foster the scientific development of junior investigators by promoting their interaction with established investigators, and (2) to allow for mixing and sharing of ideas between biochemists, molecular biologists, geneticists and systems biologists working in the field. We hope this will synergize ideas and method breakthroughs in small RNA mechanisms. |

The desire to understand the important determinants of human health and lifespan has resulted in a firestorm of work on the seven mammalian sirtuins in the past decade. The implication of sirtuins in medically important areas such as diabetes, cancer, cardiovascular dysfunction and neurodegenerative disease has catapulted them to a prominent status as potential targets for nutritional and therapeutic development. The Keystone Symposia meeting on Sirtuins in Metabolism, Aging and Disease will address fundamental questions on the regulation, biochemical, molecular and cellular mechanisms and pathways of sirtuins. The significance of sirtuin-dependent regulation of homeostasis with respect to diverse biological organisms will be discussed. Rather than focus on a specific disease, model system or technical method, this meeting will bring together a variety of experts whose primary, common trait is their innovative use of chemical and biological approaches to solve important problems in biology. From this organizing principle, the specific aims of this meeting are: (a) to assemble chemists and biologists who may not normally interact with each other; (b) to make new connections between chemists and biologists, academics and industrial scientists; and (c) to provide a forum for an open discourse of modern, cross-platform ideas.

| Résumé | Complex traits are driven by constellations of genetic and environmental factors interacting in complex ways. Genetic and environmental perturbations do not directly lead to disease but, rather, impact molecular processes that underlie physiological states associated with disease. Therefore, in order to develop a complete understanding of complex traits like disease, biological systems must be queried in a comprehensive fashion in multiple dimensions. Different data dimensions like genotype, gene expression, proteomic and metabolomic offer complementary views that are useful individually and exceptionally valuable collectively. Without mastering the large-scale molecular data that underlies pathophysiological states, without sophisticated mathematical algorithms capable of data integration, and without an appropriate informatics infrastructure to apply these algorithms and translate the results into manageable bites of information that can be consumed by physicians and patients, efforts to realize the dream of personalized medicine will be difficult to achieve.
Towards this end of realizing the promise of personalized medicine, the Keystone Symposia meeting on Complex Traits: Genomics and Computational Approaches will focus on three different areas: 1) methods for integrating diverse, large-scale, high-dimensional data with clinical phenotypes to construct predictive models (e.g., networks) of disease; 2) application of integrative biology approaches that combine large-scale molecular and clinical phenotype information to elucidate the underlying causes of disease and to prioritize drug targets and biomarkers; and 3) technological advances that will drive a dramatic explosion in data that will enable better models of disease and solutions to address the problem of how to enable others to share big data, interact with complex models and refine those models to improve our understanding of disease and associated biological processes. |

| Résumé | Apolipoprotein E4 (ApoE4) genotype is the primary risk factor for late-onset Alzheimer?s Disease. Approximately one fifth of the human population are ApoE4 carriers, yet more than 17 years after its discovery, the molecular basis by which this common allele causes neurodegeneration is still not understood. One major reason is the lack of effective communication between lipid researchers who have investigated the role of ApoE as a plasma lipid transport protein, and neuroscientists who are unfamiliar with the complex biochemistry of lipid metabolism and the cell biology of ApoE receptors. This is confounded by the methodological difficulty of studying lipid metabolism and lipoprotein transport in the brain and the paucity of pioneering work tackling this problem. Yet, the solution to one of the most pressing socioeconomic problem of our times, the looming Alzheimer?s Disease epidemic, depends on our ability to dissect and understand the molecular mechanism by which ApoE4 is causing this disease. The purpose of this highly interdisciplinary meeting on ApoE, Alzheimer?s and lipoprotein biology is to bring together a critical mass of experts with complementary expertise from these different fields, but who do not normally collaborate or attend the same conferences. In this manner, we envisage this meeting serving as the main catalyst for communication and collaboration between lipid and lipoprotein researchers, neurogeneticists, cell biologists and synaptic neurophysiologists. Opportunities for interdisciplinary interactions will be significantly enhanced by the concurrent meeting on Clinical and Molecular Biology of Acute and Chronic Traumatic Encephalopathies, which will share opening keynote addresses and a fourth day of sessions with this meeting. |

| Résumé | Acute traumatic brain injury (TBI) and its long-term neurodegenerative complications (known collectively as chronic traumatic encephalopathy, or CTE) are increasingly recognized as major public health issues, especially in the context of sports-related head injury and battlefield blast (military) exposure. Awareness of these issues has risen as neurological, psychiatric and neurodegenerative phenomena are associated with professional athletes and with veterans returning from Iraqi and Afghan theaters. The physics of head injury, as well as the epidemiological and neuropsychiatric aspects of sports-related and blast-related syndromes, will be reviewed in the initial sessions of this three-day meeting. Advances in functional neuroimaging and in the rapid and convenient determination of peptides and proteins in the cerebrospinal fluid and plasma will be reviewed in terms of their possible pathobiological significance as well as their identities as possible biomarkers. Later sessions will focus on the overlap between CTE and major neurodegenerative diseases, such as Alzheimer?s Disease (AD), frontotemporal dementia and amyotrophic lateral sclerosis. CTE is associated with accumulation of aggregated proteins or protein fragments, including tau, TDP-43, the Alzheimer?s amyloid precursor protein (APP) and the APP metabolite, amyloid-b. New experimental therapeutics research targets the roles these abnormal protein structures play in injury severity and subsequent outcome. Ab-lowering medications, currently in clinical trials for AD, improve outcome following acute brain trauma in rodent models, suggesting a pathway toward potential therapeutic interventions. Because apolipoprotein E (APOE) isotype is an important determinant of outcome from acute severe as well as mild repetitive TBI, this meeting coincides in time and venue with the Keystone Symposia meeting on ApoE, Alzheimer?s and Lipoprotein Biology. Opening plenary sessions and an optional fourth day of sessions are available for those interested in learning more about basic biology and/or clinical interventions related to APOE. |

| Résumé | In recent years there have been major advances in our understanding of the mechanisms that are involved in the recognition of microbes and subsequent activation of host immune defenses. Several classes of membrane-bound and cytosolic pattern recognition receptors (PRRs) have been identified and partially characterized. These include the Toll-like receptors (TLRs), NOD-like receptors (NLRs) and RIG-I-like receptors (RLRs). Activation of PRRs leads to the production of a large array of pro-inflammatory and anti-microbial molecules that are critical for the elimination of invading pathogens and activation of adaptive immune responses. A class of cytosolic PRRs is involved in the assembly of the inflammasome, a molecular platform that mediates activation of caspase-1 and secretion of mature IL-1b and IL-18. Importantly, activation of the inflammasome is also induced by non-microbial mechanisms including endogenous molecules involved in the pathogenesis of metabolic and inflammatory diseases. In addition, there is evidence that autophagy, a cellular process that mediates recycling of intracellular components, is involved in microbial recognition and plays a key role in host defense. The initial sensing of microbes often occurs at mucosal surfaces, but the interplay between recognition of commensal vs. pathogenic microbes on the host immune system is only beginning to be uncovered. The goal of the Keystone Symposia meeting on Innate Immunity: Sensing the Microbes and Damage Signals is to gather scientists working on innate immunity to discuss cutting-edge research on the mechanisms that regulate the activation of the immune system by microbes as well as by endogenous damage signals, and to integrate such knowledge in the context of inflammation, homeostasis, host defense and disease. Opportunities for interdisciplinary interactions will be significantly enhanced by the concurrent meeting on The Microbiome, which will share a keynote address and three plenary sessions with this meeting. |

Specifically, processes including basic immune system development, host-pathogen interactions, metabolic disease and cancer are all greatly influenced by the composition and bioactivity of the microbiota that live within us. Understanding how mammalian hosts regulate their microbiota and the myriad of ways by which the microbiota can alter susceptibility to a wide range of diseases is a key opportunity to better understand numerous diseases and, subsequently, provide strategies to promote human health. | Résumé | The complete sequences of a number of plant genomes and the advent of high-throughput technologies have, in the past few years, fueled intensive research efforts to unravel the mechanisms underlying gene and genome regulation. It is clear that a host of epigenetic processes including, but not limited to, DNA methylation and various histone modifications, underlie key developmental processes as well as biotic and abiotic interactions. Non-coding RNAs have been increasingly recognized as players in gene expression and genome defense. Various types of subcellular bodies in the nucleus have been identified and associated with transcription factors, RNA processing proteins and epigenetic regulators. Despite these advances, a holistic view of gene and genome regulation at the DNA, RNA, protein, cell biological and developmental levels is still lacking. By bringing together researchers working on various aspects of genome dynamics and gene expression, this symposium on Nuclear Events in Plant Gene Expression and Signaling aims to stimulate the synthesis of an integrative view of nuclear dynamics in plant biology. |

The field of stem cell biology interfaces with many areas of basic biology including developmental biology, tissue growth and homeostasis, regeneration, cancer and aging. Unlike most meetings on stem cells, the Keystone Symposia meeting on The Life of a Stem Cell: From Birth to Death focuses on stem cells within the context of the organism. Stem cell systems are discussed with respect to the life cycle of whole organisms: gametogenesis and embryogenesis, where tissue types are established; maturity, where organ systems are maintained in homeostasis; and aging, where stem cells systems change physiologically or because of disease. However, their specific regulation, integration within pathways and regulation are still the subject of intense investigation. Currently, they are being probed through biochemical, chemical-genetic, modeling and imaging methods. Major hurdles still reflect our very simplistic understanding of both the kinetic parameters of the molecules involved as well as the complexity of the regulatory circuits of these signaling pathways. Moreover, it is still quite difficult to translate such information into downstream biologically important information. The fact that therapeutics targeting these signaling pathways are still lagging is not surprising. The Keystone Symposia meeting on Regulation of Lymphocyte Signaling will highlight different methodologies and approaches aimed at developing a better integration of complex information about signaling. This information will lead to better understanding of how the normal immune system functions and where interventions in signaling pathways can be used in diseases of the immune system. |

| Résumé | Diabetes is associated with significantly accelerated rates of several microvascular and macrovascular complications including nephropathy, neuropathy, retinopathy and atherosclerosis. Furthermore, results from both clinical trials and animal studies indicate that these complications might progress despite glycemic control, a phenomenon termed metabolic memory. Ongoing studies have identified biochemical and nuclear mechanisms, genetic determinants, epigenetic factors and non-coding RNAs in the pathogenesis of diabetic complications. Thus, the study of the key mechanisms leading to injury in target organs and failure of adaptive processes holds great promise for the identification of mechanism-targeted therapeutic approaches. These experimental approaches have been aided by tremendous advances in high-throughput genomic and epigenomic technologies, integrated systems biology and bioinformatics. Advances in the fields of stem cell therapies, induced pluripotency and regenerative medicine have provided new hope for the treatment of complications. The goal of this interdisciplinary meeting is to integrate these evolving concepts with current knowledge to facilitate a systems-level understanding of diabetic complications. |

From the vantage point of B cell immunology, however, programmed DNA alterations at the antibody gene loci are essential for efficient antibody responses. A plethora of recent findings are beginning to illuminate open questions about the precise molecular mechanisms of these diversification processes, their regulation, and how errors in these pathways lead to cancer. Furthermore, the very same class of mutator enzymes plays an underappreciated role in non-lymphoid cancers, in antiviral responses and in epigenetic reprogramming. Antibody diversification not only shapes the repertoires of the immunological memory, but also those of auto-antibodies. The Keystone Symposia meeting on Mutations, Malignancy and Memory ? Antibodies and Immunity will provide both a platform to integrate these more or less isolated fields and an opportunity to identify interfaces that could be relevant for therapeutic approaches in malignancies and autoimmune diseases. |

| Résumé | The regulation and function of NF-kB have captivated molecular and cellular biologists, as well as immunologists, pathologists and drug developers for many years. Studies of NF-kB and its regulation have taught us many fundamental lessons and principals in basic biological sciences as well as in biomedicine. Although rapidly reaching maturity, with 25 years since its discovery, the NF-kB field remains vibrant and fertile, and never ceases to amaze with new groundbreaking discoveries. The Keystone Symposia meeting on NF-kB Signaling and Biology: From Bench to Bedside will cover the basic biochemistry of NF-kB signaling; how NF-kkB accomplishes its main job as a transcription factor; the roles of the NF-kB system in immunity and inflammation; how NF-kB controls cell death, survival and metabolism; and how all of these functions go awry during disease development ? something that we have learned both from studies of genetically altered mice and from studies of human genetic defects and pathology. Finally, this meeting will teach us how we might fix and cure these problems through the pharmacological targeting of the NF-kB system. Opportunities for interdisciplinary interactions will be significantly enhanced by the concurrent meeting on Ubiquitin Signaling, which will share a keynote address and three plenary sessions with this meeting. |

| Résumé | Post-translational modifications of cellular proteins by ubiquitin and ubiquitin-like modifiers regulate a variety of functions in the cell. A crucial aspect of signaling through ubiquitin and Ubls is the presence of modular binding domains that recognize particular types of ubiquitin signals. Such molecular coupling creates an attractive ?decoding? mechanism for responding to alterations in the cellular microenvironment and regulating physiological and pathophysiological conditions. In this meeting, we will discuss the major challenges in the field including detection of endogenous ubiquitination events, functional aspects of novel types of ubiquitin chains and their recognition by selective binding domains, and high-throughput technologies for comprehensive analysis of Ub modifications. The intersection between the deregulated ubiquitin signaling, pathogenesis of human diseases and therapies targeting the Ub system will be explored. The program offers a possibility for young scientists to present their own work and engage in discussions on the emerging topics in the field. Opportunities for interdisciplinary interactions will be significantly enhanced by the concurrent meeting on NF-kB Signaling and Biology: From Bench to Bedside, which will share a keynote address and three plenary sessions with this meeting. |

| Résumé | The 2012 Gordon Conference on Craniofacial Morphogenesis and Tissue Regeneration will focus on the molecular and genetic underpinnings of craniofacial development in an evolutionary context. It will feature a range of topics, including the early migration and specification of neural crest and mesodermal populations that compose the head, the patterned growth and signaling events that produce the major craniofacial organs, and the evolution of developmental mechanisms that give rise to the diversity of craniofacial form in vertebrates. The Conference will also cover disorders that affect craniofacial development, and animal models used to investigate their pathophysiology. A major theme will be the function of stem cells in basic developmental mechanisms and as tools in tissue engineering approaches to the treatment of craniofacial disease. The Conference will bring together an array of investigators who are at the forefront of their fields within craniofacial biology. It will provide unique opportunities for junior investigators and graduate students to present their work in poster format and interact with leaders in the field. In addition, a group of poster presenters will be selected to give brief oral presentations. The collegial and interdisciplinary atmosphere of this Conference, with formal discussion sessions as well as opportunities for informal gatherings in the afternoons and evenings, will provide an excellent venue for investigators from different disciplines in craniofacial biology to brainstorm and to develop collaborations across fields. |

| Résumé | The scientific and organizational landscape that underpins drug discovery is in a state of rapid evolution. The cellular signal transduction processes and biochemical pathways that enable life are increasingly understood at the molecular level and hypotheses for disease intervention can be proposed within this rational context. Technology to enable discovery of ligands to perturb the function of biological targets continues to advance and new methods to create small molecule tools to interrogate biological processes and potentially serve as drug leads are under development. However, while industry investment in R&D has grown exponentially, approval of new medicines has remained constant for several decades. Juxtaposition of this scientific opportunity, the continuing societal need for new drugs, and a failing model for translation of basic research into new medicines provides fertile soil for new ideas and approaches.
This meeting will focus on addressing these challenges in drug discovery with an emphasis on novel biological targets, new chemical space and emerging organizational approaches to more effective translation. The goals of the meeting are:
? To explore relatively uncharted protein target class families to define new druggable biological space
? To survey promising new chemical approaches for ligand discovery
? To highlight new organizational constructs for drug discovery with an emphasis on public/private partnerships
The meeting aims to bring together medicinal chemists, disease biologists, chemical biologists, assay developers, and screeners from industry and academics, along with leaders of R&D organizations to create a dynamic exchange around the challenges facing drug discovery. |

| Résumé | Mitochondria are no longer considered ?only? as the cellular thermodynamic converter: mounting evidence implicates them as key organelles in cell signaling, proliferation, aging, disease and death. Indeed, our view of this organelle was profoundly altered with the discovery that mitochondria function within a connected network that is continuously remodeled by fusion and division events. This reality has sparked and accelerated new scientific discoveries through the intersection of different fields about the functional roles and molecular features of mitochondrial behaviors. The interaction between the cell death and mitochondrial dynamics fields is the best example of such interplay and has led to the discovery that modulators governing cell death and mitochondrial dynamics, respectively, are in fact interacting with one another. This Keystone Symposia meeting uniquely builds on this rich interdisciplinary interaction, which continues to provide novel insights into the mechanisms underlying mitochondrial behavior and mitochondrial-associated diseases. | Résumé | Mounting evidence indicates that cell death in diseases results from complex interactions between molecular mechanisms taking place within the degenerating cells (cell-autonomous) and outside the degenerating cells (non-cell autonomous). At the cross roads of these diverse signaling pathways are found the mitochondria which, through proper mitochondrial dynamics and regulated mitochondrial elimination by autophagy (i.e., mitophagy), play a crucial role in maintaining the well-being of the cells. This meeting will focus on the general mechanisms of cell death including the Bcl-2 family and caspase proteases, the role of mitochondrial dynamics and mitophagy, the usefulness of bioinformatics to unravel death-pathways, the contribution of non-cell autonomous mechanisms to cell death and the therapeutic strategies to protect against cell death. | Résumé | The 2012 Keystone Symposia meeting on HIV Vaccines will focus on basic aspects of immunology and HIV-1 virology to highlight issues that challenge the field. The genetic diversity of circulating HIV-1 variants puts extreme demands on the quality of the response a prophylactic vaccine will need to elicit. Most likely, conserved viral determinants need to be targeted and the response must be sustained over time. To facilitate a broad and open discussion around these topics, the symposium will convene expert investigators from the HIV-1 vaccine field as well as a number of basic immunologists. Aspects of natural HIV-1 infection of direct relevance to the development of a vaccine, such as viral transmission and viral escape from immune responses, will be addressed in joint sessions with the parallel meeting on Viral Immunity and Host Gene Influence. Speakers will also be invited to present in the workshops and in the oral sessions based on submitted abstracts. |

| Résumé | This meeting will focus on recent advances in viral immunity and host gene influence that have most excited the scientific community. Systems biology approaches are being increasingly applied to the exploration of host-pathogen interactions, including the impact of viral infection on innate and adaptive immunity. Accordingly, the keynote talk will be on this topic. The first full session on the next day will focus on innate immunity, a field that continues to evolve rapidly and hold great interest in the field. In the evening the audience will hear from others who have successfully applied a systems biology approach to examine virus-host interactions. The second day explores adaptive immunity, a topic that historically has been a centerpiece for this meeting, and an area that continues to move forward rapidly. On the third day, the morning session will focus on immune evasion and subversion and will illustrate how formidable the opposition can be and why vaccine development for a number of important human diseases is so challenging. Talks on persistent and chronic viral infections, a topic with clear relevance to a number of human diseases, will be covered in the evening session. The morning session of the final day will cover immune tailoring in non-lymphoid tissues; talks will illustrate how different host responses can and must be, depending upon anatomical location of the infection. The final session will cover the latest advances in the development of vaccines and therapeutics, the traditional ending for this meeting series and a reminder to attendees of the ultimate objectives.
Overall, the objective of the Keystone Symposia meeting on Viral Immunity and Host Gene Influence is to accelerate progress in understanding and manipulating anti-viral immunity to improve human health. Opportunities for interdisciplinary interactions will be significantly enhanced by the concurrent meeting on HIV Vaccines, which will share a keynote address and three plenary sessions with this meeting. We anticipate this pairing will facilitate a wider cross-fertilization of ideas and foster innovative collaborations among scientists working in vaccinology and those studying innate immune mechanisms and other viral diseases. |

| Résumé | Over the last few years neonatologists involved in neonatal neurology have focused their attention mainly on white matter disease of prematurity. Germinal matrix intra-ventricular haemorrhage (GMH-IVH) has not been extensively studied yet, although different ways of treating its complications are provided, varying from one neonatal centre to another. A sensitive reduction of its incidence has been obtained compared to a couple of decades ago but a very likely increase is about to occur due to the increased survivability of babies at lowest gestational ages, who are also the babies with highest risk of developing GMH-IVH. The meeting pits together the best researchers available in this field to discuss the different aspects of the topic: pathogenesis, ultrasound and magnetic resonance appearance, treatment of the post-haemorrhagic hydrocephalus and its impact on later neurological outcome. Furthermore, it will present possible breakthroughs in the treatment of this disease, based on studies on new experimental animal models and clinical studies on very premature neonates. |

| Résumé | Inflammation is inextricably woven into the natural history of atherosclerosis. The macrophage is integral to the pathogenesis of atherosclerosis, functioning at the intersection of inflammation and cholesterol homeostasis, making its biological properties an important topic. Consequently, macrophage biology will be a major focus of the Keystone Symposia meeting on Molecular Basis of Vascular Inflammation and Atherosclerosis. The critical contributions of innate and adaptive immunities to atherosclerosis will also be reviewed, and a session on fundamental features of the inflammatory process, including its resolution, will introduce concepts likely to influence future paradigms in the atherosclerosis field. Recently discovered factors that regulate levels of atherogenic lipoproteins, key initiators of arterial inflammation, will be presented. Other highlights include progress in genomic approaches to discover risk factors for coronary artery disease, as well as advances in imaging techniques to detect atherosclerotic plaques, cellular composition of plaques and trafficking of immune cells in vivo. The keynote address will highlight emerging roles of nuclear hormone receptors as integrators of plaque lipid metabolism and immune activity. |

| Résumé | During the past 10 to 15 years, there has been significant progress in our understanding of pancreatic islet development and islet biology. This knowledge has been complemented and accelerated by major advances in diverse fields including islet transplantation, stem cells, small molecule screening, high-throughput sequencing, epigenetics and human genetics. The integration of these divergent disciplines has allowed us to develop a clearer definition of a functional b cell and has elucidated many of the mechanisms underlying the differentiation, survival and death of the b cell. Furthermore, these studies have revealed the potential to generate new b cells through replication or neogenesis. Improvements in differentiation and transdifferentiation protocols have also illustrated the potential of using stem cells and some somatic cell types as sources of islet cells, but these studies have also revealed the challenges associated with generating functional b cells. The purpose of this Keystone Symposia meeting is to highlight the areas of islet biology that have seen the most remarkable advances and to address some of the controversies that still exist in the field, especially with regards to the generation and maintenance of functionally mature b cells for therapeutic purposes. In addition, where it is possible, comparative mouse and human studies will be discussed to illustrate if and when mouse studies can be applied to the human situation. |

| Résumé | This meeting will highlight recent advances in virus infection pathways, and in the cellular responses to viral infection. The meeting features multiple stages of the viral lifecycle analyzed using a broad array of techniques and levels of analysis. A novel feature of this meeting is that it will be held concurrently with the Keystone Symposia meeting on Frontiers in HIV Pathogenesis, Therapy and Eradication and will, therefore, focus on maximizing exchange between the HIV community and the broader community of virologists. An explicit goal is to bring together and promote potential synergies among researchers who might not otherwise interact in more specialized meetings. A classic, almost paradigmatic, example of the power of this cross-talk is the contribution of studies of influenza A virus hemagglutinin to our understanding of the HIV-1 entry process, and the subsequent contribution of both systems to our understanding of type I fusion, type II fusion and the entry processes of non-enveloped viruses. Accordingly, two common sessions will focus on the retroviral entry process and its inhibition, followed by talks featuring a broader survey of viral entry, replication, structural and systems approaches to virology, host immune evasion and therapeutics. |

Topical to the discussion of HIV eradication is the biology behind latent infection as well as models for the study of latency and pharmacological approaches to stimulate proviral gene expression. The Keystone Symposia meeting on Frontiers in HIV Pathogenesis, Therapy and Eradication will create an environment for free exchange of ideas and cutting-edge results from a range of scientists in academia and industry in the fields of HIV pathogenesis, pharmacology and eradication. Joint plenary sessions with the concurrent meeting on Cell Biology of Virus Entry, Replication and Pathogenesis will highlight common topics of interest including mechanisms of virus entry and inhibition, and will significantly enhance opportunities for interdisciplinary interactions. Leaders from the HIV replication and drug resistance fields will critically examine the state of antiviral therapy, novel inhibitors and important virus-host interactions. The field of fibrosis is emerging and there is a strong need to understand the cellular, molecular and genetic basis of fibrosis in humans and to develop animal models that reproduce and dissect this pathological process. There is also a need to identify prognostic markers of disease susceptibility, biomarkers of disease progression and improved technologies to monitor the effectiveness of new therapies. | Résumé | It has become apparent that RNA plays a major, but still largely unexplored, role in the regulation of gene expression. It is becoming increasingly evident that longer forms of non-protein-coding RNAs (ncRNAs) are involved directly in regulating gene expression and epigenetic states. These long ncRNAs appear to be functioning as an endogenous RNA regulatory system, playing a major role in the epigenetic control of differentiation and development. These observations, across various model systems, strongly suggest that long ncRNAs are more intricately involved in the regulation of gene expression than previously envisioned. The goal of the Keystone Symposia meeting on Non-Coding RNAs is to highlight the similarities and differences in various model systems to better understand the role of long ncRNAs in the regulation of genomes. This is an entirely new emerging field of study that is on the forefront of modern biology. Opportunities for interdisciplinary interactions will be significantly enhanced by the concurrent meeting on Eukaryotic Transcription, which will share a plenary session with this meeting. |

| Résumé | Gene expression in eukaryotes is regulated primarily at the level of transcription. Biochemical, genetic and molecular genetic analyses of a handful of genes have provided a basic understanding of transcription. Advances in proteomics have rapidly expanded the repertoire of proteins and protein-protein interactions that participate in gene expression, and genome-wide analyses of protein distributions across genomes and gene expression are expanding and challenging our knowledge. For example, RNA polymerase II is found concentrated at the promoters of thousands of genes, causing much interest in the field to shift focus to the role of elongation in regulating gene expression. The functions of vast noncoding, transcribed regions of the genome are largely unknown. HDACs, thought to function primarily as repressors, tend to be associated with highly transcribed genes. This meeting will explore new and exciting aspects of eukaryotic gene regulation emerging from genomic, proteomic and other high-throughput techniques. |

| Résumé | A fundamental goal of neuroscience is to understand the molecular, cellular and activity-based mechanisms that control the formation and maintenance of neural circuits and determine how these mechanisms become compromised in neurodevelopmental, psychiatric and neurodegenerative disorders. Over the past two decades, molecular neuroscientists have identified key molecules and mechanisms that underlie synapse development, activity and stability. Meanwhile, the study of neuronal circuits has been revolutionized by new methods to visualize and map circuits in living animals, as well as the development of approaches to control neuronal activity using optical approaches (so called optogenetics). Finally, disease researchers have identified genes associated with neurodevelopmental and psychiatric disorders and neurodegenerative diseases. Animal models of these diseases are proving useful to understand how dysfunction of affected genes and proteins contributes to disease pathology. Although these fields are working on the same processes, no small highly interactive meeting brings these three groups together. The Keystone Symposia meeting on Synapses and Circuits: From Formation to Disease will address this need by bringing together leaders working on synapse development and function, circuit structure and function and the study of brain disease. We anticipate that mutually beneficial insights will emerge from discussions at this meeting. |

Current efforts focus on the application of stem and progenitor cells, bioactive factors and biomaterial scaffolds in transplantation procedures to affect regenerative therapies. Breakthroughs in bladder and tracheal reconstruction and highly promising approaches for solid organ regeneration as well as musculoskeletal and nerve repair have been achieved. Effective cell sourcing, optimal assembly and fabrication of tissue-derived and synthetic biomaterial scaffolds, and targeted regulation of therapeutic and immunomodulatory functions of stem cells represent some of the current challenges in this field. In this Keystone Symposia meeting on Regenerative Tissue Engineering and Transplantation, exciting technological and intellectual advances in the field will be synthesized by expert scientists, engineers and clinicians to assess and expand the landscape in regenerative engineering and transplantation. Opportunities for interdisciplinary interactions will be significantly enhanced by the concurrent meeting on Mechanisms of Whole Organ Regeneration, which will share an opening and closing keynote address as well as two plenary sessions with this meeting. More specifically, the pairing of these two meetings represents a highly unusual opportunity for cross-talk between the more applications-driven field of tissue engineering and regenerative medicine, and the very basic research being explored in the field of whole organ regeneration. |

This meeting aims to delineate the molecular and cellular biological basis of regeneration by highlighting, comparing and contrasting the differences and similarities in regenerative capabilities and mechanisms among diverse animal species. How these biological findings may inform regenerative medicine and stem-cell biology and therapeutics will be considered and discussed. | Résumé | According to the World Health Organization, over 25% of new tuberculosis cases in some subnational regions are multi-drug-resistant TB (MDR-TB) and, as of March 2010, 58 countries had reported XDR-TB (TB resistant to the major classes of both first- and second-line drugs). The Keystone Symposia meeting on Drug Resistance and Persistence in Tuberculosis aims to bring together basic and clinical researchers and public health experts from diverse backgrounds to explore the following: 1) the fundamental biology underlying genetic and phenotypic drug-resistance in TB; 2) progress in expanding capacity in high-burden settings to perform drug susceptibility testing; 3) identification of novel drug targets; 4) innovative strategies for drug discovery; and 5) advances in developing multi-drug regimens that will be effective against currently drug-resistant as well as drug-sensitive strains of M. tuberculosis. Investigators from outside the TB field will be included to provide new perspectives and stimulate fresh approaches for tackling this global public health priority. |

| Résumé | The Gordon Research Conference on NOX Family NADPH Oxidases meets biannually, alternating between US and European sites so as to attract a broad range of international scientists active in this rapidly growing field of biomedical research. The 2012 meeting will also include the first NOX Gordon Research Seminar for research trainees. The NOX GRC will feature cutting-edge unpublished work presented by experts across the NOX field, whereas the NOX GRS will be geared specifically to the interests and needs of young research trainees in the field, primarily graduate students and post-doctoral fellows. NADPH oxidases of the NOX family have in recent years been shown to comprise the predominant source of reactive oxygen species (ROS) produced by most types of cells. It is abundantly clear that the science in this field is moving very rapidly, with many exciting advances in understanding of the basic biochemistry and cell biology of the NOX family of enzymes, particularly the contrasting properties and functional roles of the seven discrete isoforms comprising the family. Moreover, NOX-generated ROS are directly involved in a wide range of both physiologic and pathologic processes. Many linkages between NOX enzymes and disease pathogenesis are being elucidated and this knowledge is dramatically accelerating advances in the identification of new and often unexpected therapeutic targets and development of disease-modifying pharmacologic agents. The NOX Gordon Conference program has been clearly established as the top forum for the presentation and discussion of the latest advances in this field. Thus, the 2012 NOX GRC/GRS meetings promise to be timely, exciting, and paradigm-challenging. The program for the meeting has been developed around the theme of "NOX Biology and its Translation to Human Disease and Therapy." The preliminary conference program comprises nine scientific sessions, four with basic themes and five with a disease-oriented translational emphasis, including one session devoted entirely to recent exciting advances in the development and testing of small-molecule NOX inhibitors. Each session features a discussion leader and three to four speakers, plus a few young investigator presentations to be selected from among an anticipated ~100 poster abstracts. There will be ample time for vigorous discussion after each talk. The GRS program will feature speakers selected from among trainee abstracts, as well as career development components and mentoring by senior scientists. Attracting a diverse group of participants is a major priority, as reflected for example in the high prevalence of women scientists (35%) listed in the preliminary GRC program. |

| Résumé | Oceans interact with humans in both positive and negative ways and this interaction has become the foundation for an emerging "metadiscipline" called Oceans and Human Health. The 2012 Gordon Research Conference on Oceans and Human Health (OHH) will address several of the many disciplines comprising OHH that include oceanography, waterborne and seafood borne diseases, harmful algal blooms, epidemiology, comparative animal physiology, natural products and synthetic organic chemistry, pharmacology, toxicology, social sciences, engineering, and other ocean-related areas. This conference will feature sessions on seafood, aquaculture, omics, natural disasters, climate change, forecasting, changes in fecal indicator detection, marine human pathogens, emerging chemicals of concern and communications, all as they relate to human health. It will have a session on the Deepwater Horizon to update the conferees on what research and restoration has transpired since the April 2010 oil blowout in Macondo Canyon. And finally, there will be discussions on a developing OHH initiative by the European Science Foundation's Marine Board and the future of OHH in the U.S. As is the case with all GRCs, this conference will assemble experts in the several session areas who will present current information to junior scientists and graduate students in a very relaxed atmosphere. In addition to the formal presentations, poster presentations will provide opportunities for "one-on-one" discussions about OHH topics. |

Les Diablerets provides an intimate setting in the Swiss mountains for the 2012 GRC on Immunochemistry and Immunobiology, where a maximum of 150 participants will exchange ideas and discuss the latest advances in the field. In contrast to many conferences that focus on specialized Immunology topics, this 2012 GRC on Immunochemistry and Immunobiology will feature broad coverage of both innate and adaptive immunity, and will span the full spectrum of basic mechanisms to applied research. The meeting will be divided into nine half-day sessions, with approximately 36 invited internationally recognized speakers. Additional slots will be allotted for short oral presentations selected from submitted abstracts. There will also be poster sessions and many opportunities for informal interactions among participants throughout the week. | Résumé | The 2012 Cardiac Regulatory Mechanisms Gordon Research Conference is one of the premier meetings for scientists interested in cellular and molecular mechanisms regulating the normal and diseased heart. In addition to the role of Ca in excitation/contraction coupling and contractility that formed the original basis for this meeting, the 2012 meeting will integrate studies at the nexus of Ca, ion channels, redox and kinase signaling, and mitochondria in regulating cardiac cell function and diseases such as sudden arrhythmias, hypertrophy and heart failure. Emerging data has shown an important role for redox and kinase regulation of ion channels and transporters. Compelling data also show cross-talk between kinase and redox signaling. Mitochondria are involved in ion signaling, redox regulation, metabolism and cell death and play an important role in regulating excitation contraction coupling and cardiac function. The meeting will report on mechanisms by which these pathways function and interact. Topics include novel models for studying cardiovascular disease, new mechanisms and targets for redox signaling, signaling via contractile proteins, the intersection of calcium, signaling and hypertrophy, mitochondrial regulation of Ca signaling and cardiac function, the importance of location in signaling, ion channels and arrhythmias and novels aspects of Ca dynamics. The meeting will cover basic mechanism as well as perspectives on translating these findings into a better understanding of disease mechanisms, diagnosis, prevention and treatment. The goal of the meeting is to bring together scientists who can critically evaluate developments at the forefront of cardiovascular research. This is accomplished by including established as well as early career research scientists from the fields of physiology, pharmacology, molecular biology, biochemistry, biophysics and bioengineering, along with physician/scientists with an appreciation for the therapeutic implications of cardiovascular research. Based on past experience, we can guarantee an atmosphere of intense and lively discussions where all participants contribute and learn. |

| Résumé | Fragile X syndrome (FXS) is the most common form of inherited intellectual deficiency and the most common identifiable single gene cause of autism, affecting 1 in 5,000 males and a lesser number of females. Autism spectrum disorders (ASD) occur in up to 2/3 of males and 1/3 of females with FXS. The Fragile X gene (FMR1) was cloned in 1991 and since then a large field has grown with roughly one hundred labs using techniques from biochemistry through genetics to model organisms to elucidate the functions of the FMR1 protein (FMRP). FMR1 gene homologs have been found in Drosophila and mice, and useful null mutant models generated in both. FMRP has been found to be involved in the regulation of specific messenger RNA transport, localization and expression in neurons. Briefly put, FMRP is thought to bind and help transport a crucial subset of messages to the sites of neural action (the synapses). At those synapses, FMRP is a critical switch that mediates changes in local protein expression in response to neural activity - in effect causing the synapses to strengthen or weaken as required in response to experience. When FMRP is reduced or missing in an affected person, the synapses are much less able to change with experience and learning is greatly reduced. More recently, specific signaling mechanisms have been proposed to mediate this control at the synapse, via a specific set of Gq-linked (neurotransmitter) receptors, including group I mGluRs (metabotropic glutamate receptors). It has become clear that in addition to clinical overlap between FXS and ASD, there is likely substantial overlap in the molecular pathology of the two disorders. As such, molecular defects known to cause ASD may involve other proteins in the signaling pathways that are regulated by or regulate FMRP activity, may involve proteins for which synaptic translation is regulated by FMRP, or may involve defects in neurotransmitter systems shown to be dysregulated in FXS models. Molecules aimed at targets in pathways that are dysregulated in the absence of FMRP are now being developed and tested in academic laboratories and through the pharmaceutical industry, in order to offer effective drug therapies for affected patients with FXS. It is expected that many of these targeted treatments will have therapeutic overlap in subsets of individuals with ASD. This conference will bring together leading scientists and clinicians in the Fragile X and ASD fields. Topics will include molecular genetics, model system characterizations, synaptic signaling and function studies, small molecule trials and clinical reports. |

The conference will place special emphasis on human and animal diseases such as malaria, toxoplasmosis, trypanosomiasis, leishmaniasis, trichomoniasis, giardiasis, entamoebiasis, schistosomiasis and gastrointestinal nematode infection. The program will feature a wide range of topics, such as host and parasite co-evolution, parasite cell and developmental biology, pathogenesis and virulence, host invasion, metabolic interaction, immune response and immune evasion, and drug action and resistance. The aim of the meeting is to bring together scientists from a variety of disciplines in order to define the current state of this dynamic field and to discuss and set future research agendas. This GRC conference offers the unique opportunity for junior investigators and graduate students to get together and exchange ideas with leaders in the field in a friendly and relaxed atmosphere at the Salve Regina University Campus, Newport. Some submitted abstracts will be selected for short talks. Students and post-docs are strongly encouraged to apply. We are making a special effort to obtain funds to provide individuals from endemic countries with travel awards. |

| Résumé | The 12th biennial Gordon Conference on Intermediate Filaments (IF) will focus on exciting topics of interest to cell biologists, researchers engaged in biomedical research, physicists and engineers in this rapidly growing field of research. Emphasis will be placed on the structure and function of the large number of proteins that polymerize into dynamic cytoskeletal and nucleoskeletal networks. IF proteins are encoded by one of the largest families of genes in the human genome giving rise to over 70 proteins. These genes are expressed in a developmentally regulated fashion resulting in a remarkable array of both cytoskeletal and nucleoskeletal networks. Together with their binding partners, cytoskeletal IF including the epithelial cell keratins, the mesenchymal cell IF proteins such as vimentin, desmin and GFAP as well as the different types of neural IF, are rapidly becoming recognized as essential elements in the differentiation of cells and tissues. These IF cytoskeletal systems are major factors in the development and maintenance of cell shape; they are also determinants of the mechanical integrity of cells due to their unusual strain stiffening properties. Recent studies have revealed a remarkable array of additional functions for cytoskeletal IF including mechano- and chemo- sensing, regulation of major signal transduction pathways, cell migration, the positioning and anchorage of organelles such as mitochondria, the epithelial/ mesenchymal transition, and cell/cell and cell/substrate adhesion. IF also act as central elements in the crosstalk among the different cytoskeletal systems including microtubules and actin. Within the nucleus, the IF proteins known as the nuclear lamins are the major structural components of the nuclear lamina/envelope and they play essential roles in the shape, size and mechanical properties of the nucleus. More recently the lamins have been recognized as central determinants of the molecular architecture of the interphase nucleus, the organization of chromosomes, the positioning and expression of genes, DNA replication and the disassembly and reassembly of the nucleus during cell division. The remarkable discoveries of hundreds of mutations in IF genes giving rise to a plethora of human diseases have attracted the attention of broad audiences of basic scientists and physicians. Theetiology and manifestations of these diseases are wide-ranging, including premature aging diseases such as progeria, cardiomyopathies, neurodegenerative disorders, numerous forms of muscular dystrophy, as well as blistering diseases of the epidermis, amongst many others. In other diseases abnormal aggregates of IF are now being recognized as factors in motor neuron diseases, Alzheimer's disease and giant axonal neuropathy. This Gordon Research Conference is the world's largest forum for fostering interactions among IF and other interested researchers. The Conference provides an unprecedented opportunity to learn about the latest cutting edge developments in the functions of IF. This GRC has always fostered a free exchange of ideas, tools and reagents, and has facilitated the establishment of many productive interdisciplinary collaborations. The attendance of young scientists is strongly encouraged. In support of this the GRC Board of Trustees has provided resources to inaugurate the first IF Gordon Research Seminar which will precede the Conference and will be organized by graduate students and postdoctoral fellows. All applicants are invited to present posters describing their latest research and a number of platform presentations will be chosen from the submitted abstracts in order to ensure inclusion of the latest breakthroughs during the formal sessions. Significant time for discussion will be scheduled into the sessions to promote the lively exchange of ideas and to consider alternate interpretations of data presented. The schedule will also allow ample free time in the afternoons and after the last evening session to foster the informal interactions that have been the hallmark of the IF Gordon Conferences. |

| Résumé | Dysregulated hemostasis is a major cause of human disease and death. The Gordon Research Conference on Hemostasis is considered the premiere small format meeting on the topics of blood coagulation, platelet biology, anticoagulant pathways and associated diseases. First organized by hemostasis pioneers Earl W. Davie and Oscar Ratnoff in 1973, the meeting was held yearly until 1992 and biennially since then. The Hemostasis Gordon Conference brings together scientists interested in the biology, biochemistry and genetics of platelets and coagulation proteins, physicians involved in the treatment of bleeding and thrombotic diseases and investigators from small and large pharmaceutical companies developing new therapies for the diseases of hemostasis. The Hemostasis Gordon Conference enjoys an international reputation for being the forum at which cutting-edge research is presented and major advances in the field are unveiled. The next conference will be held on July 22-27, 2012 at the Waterville Valley Conference Center, Waterville Valley, New Hampshire. The site is regarded as a top venue providing comfortable meeting and housing facilities in an inviting location. This Conference Center, surrounded by the White Mountain National Forest provides over 15,000 square feet of flexible, highly functional space. Recreational facilities include ready access to biking, hiking, golf, tennis, boating, chair-lift rides, an athletic center and the Waterville Valley Arts Center. All accommodations, dining hall, poster areas and meeting rooms are accessible to handicapped persons. Waterville Valley is two hours north of Boston and a bit more than an hour from Manchester, New Hampshire. Travel to the site is convenient for attendees from the US and overseas alike. It is the goal of the conference organizers that nearly every person attending the conference participates either as a speaker, discussion leader, or poster presenter. At Waterville Valley, the venue for the 2012 Conference, there is space available for 30-40 posters per session, and four poster sessions will be scheduled. The conference Vice-Chair, Andrew Weyrich, will be responsible for organizing the posters into sessions that are related to the meeting agenda and for the selection of top posters for presentation as a series of short talks in a Hot Topics session. Afternoons will be open for informal discussion among attendees and leisure activities aimed at fostering further interactions and intellectual exchange - traditionally a central feature of all Gordon Conferences. Poster sessions will be held before dinner on four of the five days. These sessions provide a dedicated, informal forum for interaction between junior and more senior scientists in the field. All applicants who are not invited speakers are encouraged to submit an abstract for presentation at one of the four poster sessions (Monday-Thursday) and for consideration to be chosen for a short oral presentation at the Hot Topics session on Wednesday evening. |

In most cases, drug resistance is investigated as a disease specific endeavor, with no centralized, cross-disciplinary network in the world, limiting intellectual progress and breakthroughs. Clinicians and researchers are often unaware of the progress, resources and parallels in other diseases. However, it is increasingly clear that close parallels can often be drawn between resistance seen in one disease state and resistance seen within another. Cross-disciplinary collaboration and discussion are permitting a better understanding of resistance and may soon result in real clinical benefits. The new Gordon Research Conference entitled "Drug Resistance" is devoted to recognizing the parallels among quickly evolving diseases. | Résumé | The Gordon Research Conference on Musculoskeletal Biology and Bioengineering (previously titled Bioengineering and Orthopaedic Sciences) has been the premier forum for presentation and discussion of new and unpublished information in the field, and has consistently led to new insights, new interactions, new collaborations, and new research directions. The study of the musculoskeletal system encompasses a number of interdisciplinary fields, particularly biology and bioengineering, and has ultimate applications in clinical areas including orthopaedic surgery, rheumatology, and radiology. The theme of the planned 2012 conference is "Musculoskeletal Science: Bedside to Bench to Bedside," and the meeting will consist of 9 separate sessions, consisting of invited speakers who are experts in their field and selected attendees at various levels of career development. The format of the planned conference remains essentially unchanged from previous years, with ample time for formal and informal discussions. Our unique focus in 2012 is to facilitate the physician-scientist interaction to bring new ideas and treatments to improve the lives of patients. This is exemplified by our session on Translational Studies on Enhancing Soft Tissue Healing, with both physicians and scientists lecturing together on cutting edge solutions and investigations into clinical problems. Some of the chief issues are specific to individual tissues, including bone, articular cartilage, intra-articular ligament, meniscus, and both rotator cuff and flexor tendons, as well as other multi-tissue skeletal structures. A number of key scientific and engineering topics are relevant to fabrication and manipulation of these tissues and organs. The conference will examine many of these topics, including biomaterial fabrication, stem cells, biomarkers, skeletal development, growth and regeneration, and translation of basic research to clinical practice. In addition, the conference also includes several high-profile research areas highlighting the close collaboration required between clinician-scientists and PhDs to improve patients' health outcomes. |
