Keystone Symposia: Antibodies as Drugs - Translating Molecules into Treatments

Antibodies carry an enormous potential in prophylaxis and treatment of human disease. Antibody therapies may be divided into two distinct but highly related disciplines. First, antibodies can be elicited de novo using vaccines. Our understanding of what constitutes an effective antibody response at the molecular level and the science of eliciting such antibodies in vivo is progressing rapidly. Second, isolated antibodies represent powerful drugs when developed as biopharmaceuticals, as demonstrated by the ever-expanding list of approved molecules as well as by the large number of antibody products in development in a wide range of therapeutic areas. The antibody landscape is changing as the field has moved from empirical to knowledge-based, designer approaches for which understanding of mechanisms of action at the molecular level is critical. Translating such basic knowledge into novel products and applications requires scientists from many disciplines including structural and cellular immunology, biology, engineering, chemistry and medicine to interact closely. Consequently, the field of antibody-based vaccines and therapeutics has become an interface area where experts meet and work together. The 2018 Keystone Symposia antibodies conference aims to provide a platform for knowledge-sharing from the molecular to the systems level to navigate the newest developments and insights to prevent, treat and cure major human diseases.

Keystone Symposia - Endoderm Development and Disease: Cross-Organ Comparison and Interplay

Organs do not function in isolation. In contrast to the majority of existing meetings that focus on individual organs (lung, pancreas, intestines, etc.), or individual pathways (FGF, WNT, Hippo, etc.), this conference offers a unique cross-organ perspective on development, homeostasis, repair, regeneration and pathogenesis. The endoderm is an ideal setting as it gives rise to the majority of our internal organs such as the thyroid, thymus, lungs, liver, pancreas, esophagus, stomach, intestine and prostate, etc. Associated with these organs is a wide spectrum of diseases, including asthma, diabetes, fibrosis, obesity, infections and cancer. This conference aims to achieve the following goals: 1) Synthesize similarities and differences of mechanisms of specification, morphogenesis, maturation, regeneration and pathogenesis across organs; 2) Use the simplicity of the developing organs to uncover complex disease mechanisms; 3) Investigate organ development and function in the context of other organs, the immune system and the nervous system; 4) Consider organ physiology and metabolism under the influence of the environment: bacteria, viruses, allergen, pollutants and circadian control; and 5) Share successful technologies across organ systems. Such a cross-sectional perspective brings together researchers across scientific disciplines who do not normally meet. The meeting will link development with physiology and help to establish a community of the next generation of researchers who will approach their scientific investigation with a broad, multi-systems view.

Keystone Symposia - Immunological Memory: Innate, Adaptive and Beyond

This symposium will primarily focus on integrating current knowledge concerning the cellular and molecular regulation of memory T and B cell responses with their role mediating protection against infections. In addition to the significant impact these cell populations have on vaccination, emphasis will be placed on their therapeutic and pathogenic roles in cancer, autoimmunity and chronic infection. The innate immune response has a profound influence on the regulation of effector and memory T and B cell responses, and recent discoveries in this area bring forth a greater understanding of the major signals that govern both the formation and the long-term maintenance of these populations. These conceptual advancements will be a major part of the symposium. More recently, it has become clear that there is marked phenotypic and functional heterogeneity of T cell responses; how this affects immunological protection will be discussed. Finally, this symposium will include discussion of recent advances in assessing memory T and B cell responses in a sensitive manner in human and non-human primates, as well as murine systems, to enable better elucidation of the state of the immune response during infection and vaccination, which will lead to more rationale vaccine design. The symposium is being paired with "Aging, Inflammation and Immunity."

Keystone Symposia - Aging, Inflammation and Immunity

The world’s aging population and problems with morbidity have been increasing rapidly. Decreases in immune function and increases in chronic inflammation with age are a major contributor to age-related diseases. This Keystone Symposia conference will provide a venue for researchers from around the world to discuss current findings and fundamental research to understand the mechanisms underlying declining immune function, increasing chronic inflammation, and possible interventions. The meeting will address essential aspects of aging/longevity research, with particular emphasis on the immune system and inflammation including effects of immunosenescence on cancer, autoimmunity, vaccine response and infectious disease. Longitudinal studies on aging and immunity, frailty and effects on mortality, quality of life and survival will also be covered as well as biomarkers of the aging immune system, mechanisms for generating chronic inflammation including the SASP, telomeres and the microbiome, regulation of aging and age-related immune function, and therapeutic approaches to slow aging and/or improve the immune system. The adaptive, innate and mucosal immune systems will be covered as these systems have a significant impact on human aging. Questions to be addressed include: 1) What changes in the immune system contribute to the observed decline in immune function and increase in the chronic inflammatory state? 2) How do diseases of aging or decreased immunity develop, and what can be done to prevent or reverse them? To address these questions, the symposium will gather investigators from interdisciplinary areas on aging, inflammation and immunology and will focus first on the human and secondly the murine immune systems. This meeting is paired with "Immunological Memory: Innate, Adaptive and Beyond."

Cells have evolved complex mechanisms to detect and respond to infection. We are only beginning to uncover the cellular mechanisms that engage invading pathogens to restrict infection. While bacteria, viruses and parasites are seemingly different, emerging data suggests that there is a convergence of cellular pathways that target these invading pathogens, which these pathogens have developed subversive strategies to evade. This conference will focus on host processes at the forefront of this battle and how pathogens are targeted. From the initial stages of non-self-recognition to the effector pathways engaged, these diverse pathogens are both manipulated and manipulating the system. Cutting-edge technologies are driving our deeper understanding of how core biological processes can be regulated in the favor of the host, or pathogen, and how these new mechanisms might be harnessed to treat diverse diseases. This meeting focuses on intrinsic mechanisms of defense across pathogens, bringing together diverse areas of expertise showcasing the convergence of pathways engaged.

Keystone Symposia - Cancer Immunotherapy: Combinations

By 2025, most patients with cancer will receive immunotherapy as part of their treatment regimen. This symposium will cover the most critical topics integral to this vision: How can we integrate, sequence or combine immunotherapy with standard-of-care cytotoxic therapy, radiotherapy and targeted therapies? Which preclinical models are best predictive of combination approaches? How do we combine immune agonists as well as molecules inhibiting immune-suppressive pathways with checkpoint inhibitors? What are the rational combinations for cancer vaccines and viro-therapy? Which combinatorial approaches could prevent or treat adaptive mechanisms of resistance? Most immunotherapies are being developed in isolation, or being tested as single agents in preclinical models or in clinical studies. This conference helps to address the gaps in knowledge of how and when to combine therapies, and how to integrate immunotherapy into current standard-of-care or novel targeted therapy approaches – both preclinical and clinical. Overall, the objectives of this meeting are to: 1) Elucidate rational combinations for immunotherapy; 2) Discuss preclinical and other models to inform combinatorial approaches; 3) Understand the relevance for biomarker approaches; and 4) Provide insights into the latest preclinical and clinical data for immunotherapy combinations.

Keystone Symposia: The Resolution of Inflammation in Health and Disease

Résumé:
Inflammation and its resolution are critical to effective host defense and tissue homeostasis. There is a steady increase in the recognition that the effective resolution of inflammation is dynamically regulated through cellular responses to endogenously generated mediators. Ineffective resolution of inflammatory responses likely contributes to the pathogenesis of numerous prevalent conditions including atherosclerosis, arthritis and diabetes. Unresolved chronic inflammation may lead to fibrosis and eventual organ failure. Whereas anti-inflammatory therapeutic approaches are conventional, a growing consensus based on experimental data suggests that an alternative approach may be to mimic agonism of resolution. The aim of this meeting is to enhance our understanding of the molecular mechanisms which underpin the effective resolution of inflammation and how these may fail in aging and disease. Specifically, it seeks to: 1) Encourage dialog between those working in areas where the potential of mimicry of resolution of inflammation might play an under-appreciated role; 2) Consolidate data from diverse mediators that promote resolution; and 3) Contribute to the education and training of graduate students, fellows and faculty. These discussions will provide a multidisciplinary community with insight into physiologic processes underpinning resolution and potential mimicry of such processes for therapeutic gain in pathological conditions.

Microbiologists and immunologists will again come together to provide different viewpoints on the same problems in joint sessions. While maintaining continuity in topics, the meeting concept will be evolved by introducing novel sessions on bacterial defenses against innate immunity, an area prominently covered by abstract submissions in the 2016 meeting, and on fungus host interactions, an under-researched area of great medical importance.

Emerging and re-emerging viruses have the potential to cause high morbidity and mortality and range from localized outbreaks to epidemics. Due to their emerging nature, most aspects of the biology and infectious potential of these viruses are poorly understood. Our continuing struggle to respond to a procession of pandemics including SARS, avian influenza, MERS, Ebola and more recently Zika, highlights key gaps in our knowledge and should serve to motivate our re-thinking on how we can better prepare and deal with future unknown viral threats. This meeting will focus on important areas such as surveillance, diagnostics and countermeasures and other important advances in new technologies and how they are being applied to research. Furthermore, we will discuss how to facilitate the translation of research, data and candidate treatments through the development pipeline in a timely and cost-effective manner. The key themes to be covered include the need to understand why zoonotic diseases matter, their association with agriculture, the importance of surveillance and early detection, and the difficulties of dealing with diseases that involve both medical and veterinary communities. The conference will bring together experts in virology, immunology, vaccinology, epidemiology with those that seek to transfer knowledge between these groups, veterinarians and industry and government. Further, this meeting would bring together individuals involved in the control of these diseases in government and non-government organizations, as well as people involved in study of zoonosis and countermeasures. The creation of global networks and sharing of information will ensure that we are better prepared for future outbreaks.

Infectious diseases cause substantial morbidity and mortality in the developing world. The medicines available today are often inconvenient to use or result in side effects, and many are being lost to emerging drug resistance. There remains an urgent need to discover and develop the next generation of transformative medicines that are more efficacious, more highly targeted, and have a better safety profile. This will only be achieved through the application of state-of-the-art drug discovery tools, technologies and approaches. This meeting will focus on diseases caused by parasitic organisms and bacteria, including malaria, neglected tropical diseases, diarrheal disease and tuberculosis. The goals of this meeting are to improve understanding of the disease burden and challenges faced in developing new therapies, to highlight examples of progress towards new drug candidates, and to foster communication and collaboration among communities of researchers working in global health across the spectrum from research to the clinic. The meeting will be of interest to basic scientists as well as drug discovery and development experts wanting to gain a better understanding of the challenges and opportunities in global health.

Keystone Symposia: Role of the Genital Tract Microbiome in Sexual and Reproductive Health

Résumé:
Bacteria and fungi residing in the female genital tract have been associated with elevated risks of HIV acquisition and transmission, pelvic inflammatory disease, miscarriage, preterm birth, and invasive maternal and neonatal infections. The increased availability of high throughput genomic testing since the turn of the century has revealed a more detailed picture of these organisms than was possible when evaluation depended on microscopy and culture. The interrelationships between sexually transmitted infections, vaginal dysbiosis, vulvovaginal candidiasis, and vaginal pathobiont carriage are being elucidated, and their effects on the cervicovaginal mucosal barrier and immune system are being characterized. The mechanisms that may lead to adverse outcomes are being unraveled, and an increasing number of interventions are in clinical trials. In this meeting, the current understanding of female and male genital tract microbiology and immunology, including functional microbiology and biofilms, will be presented and knowledge gaps identified. Potential mechanisms leading to adverse outcomes, and a variety of potential prevention and/or treatment interventions, will be presented and discussed. The exponential progress made in recent years will hopefully lead to efficacious public health interventions to reduce the high prevalence of adverse sexual and reproductive health outcomes in women, especially in resource-poor settings.
