Cancer Genetics Course 2018

The field of cancer genomics and recent technological advances are shaping how we diagnose and treat cancer. This skills based course will teach the core concepts of risk stratification using family history as a tool to enable patients to access appropriate management strategies. It is a course designed for all healthcare professionals working in primary care and specialist settings including oncology, breast care, gynae-oncology, gastroenterology and screening services. Participants will be equipped with the basics of cancer genetic counselling and sessions will include approaches to genetic testing, management of hereditary cancers and consent taking.

What is Oncology Clinical Experience? Oncology Clinical Experience is a hospital based oncology training course and combines theoretical teaching with practical example. Physician taught lectures are combined with visits to clinics and delegates have the opportunity to interact with patients in small group sessions. Oncology Clinical Experience offers rapid and progressive face-to-face training taught by clinician experts and the care team.  This course thoroughly equips attendees with theoretical and practical knowledge to discuss Oncology and its treatment confidently and credibly. Participants are provided with an overview of the spectrum of malignant disease and there is unique opportunity for delegates to experience – first hand – the assessment and treatment options utilised within the overall management of patients with malignant disease. Keystone Symposia: T Cell Dysfunction, Cancer and Infection

T cell exhaustion is an acquired state of T cell dysfunction, associated with reduced T cell function, sustained expression of inhibitory receptors and a transcriptional state that is distinct from memory and effector T cells. T cell exhaustion is now recognized as a defining feature not only of chronic infections such as HIV, malaria and tuberculosis, but also of cancer. As a result, the search for effective approaches to reverse T cell exhaustion is the focus of immense therapeutic development. However, the similarities and differences between T cell dysfunction arising in chronic infection and tumors are not fully understood. Moreover, opportunities to identify new therapeutic approaches to reverse dysfunction based on parallels between the two disease states remain to be explored. This meeting will focus on the comparative biology and clinical therapeutics of T cell dysfunction in chronic viral infection and cancer. Drawing on leaders in fundamental T cell biology, computational science and clinical investigation, it will focus on common features and distinctive aspects of T cell dysfunction in cancer and chronic infection. The meeting will have broad relevance for T cell biology, tumor immunity, infectious disease and therapeutic development.

This course reviews clinically important information regarding the management of cardiac and vascular diseases affecting patients with cancer and cancer survivors. Keystone Symposia - Cancer Epigenetics: New Mechanisms, New Therapies

The field of epigenetics has recently emerged as an essential component of oncology research, with altered epigenetic modifications considered to be critical hallmarks of cancers. Unlike mutations, epigenetic aberrations are reversible and as a result, epigenetic therapies hold great promise as anti-cancer agents. For these novel therapies to succeed, a key challenge is to fully understand the nature and the mechanisms that lead to epigenetic changes in cancer. Another essential challenge is to make sense of the avalanche of data flowing from increasingly powerful "omics-based" technologies. Despite these challenges, never has the potential for scientific and clinical breakthroughs in this field been greater. Bringing together both the academic and the pharmaceutical communities, this Keystone Symposia conference will focus on: 1) The newest findings concerning the mechanisms by which epigenetic pathways are established and maintained in normal and stem cells, and how these go awry during aging and cancer, with a special emphasis on histone modifications, chromatin remodeling and the unexpected increased number of DNA modifications ((hydroxy)methylcytosine and methyladenine); 2) The growing roles of noncoding RNAs in cancer and the emergence of the new and exciting realm of RNA epigenetics; 3) Use of new omics technologies to interrogate altered epigenetic landscapes in tumors and assess their clinical implications; and 4) Novel and promising therapeutic drugs aimed at reversing specific epigenetic alterations. Ultimately, the meeting will provide great opportunities for senior and junior scientists to meet the leaders in the field from both academia and industry, hence forging exciting interdisciplinary interactions and creating potential new collaborations in the booming area of cancer epigenetics.

3rd DNA Replication/Repair Structures and Cancer - Fusion Conferences

The remarkable stability of the human genome is lost in cancer cells due to the failure of efficient and accurate repair in the context of oncogene-induced replication stress and elevated transcription. DNA replication is furthermore emerging as a surprisingly fragile and complex process requiring fork protection and restart, template-switching, R-loop resolution, gap-filling, and repair. Unresolved replication and repair intermediates signal apoptosis. The synthetic lethality and essentiality resulting from replication-repair stresses thus suggest repair inhibitors as tools to control pathway selection and damage outcomes and to design advanced therapeutics. The 3rd DNA Replication/Repair Structures & Cancer conference in February 2018 will focus on structural and mechanistic insights into dynamic protein, chromatin, DNA and RNA complexes acting in DNA replication and repair events relevant to cancer. Conference talks and discussions will center on developing an actionable mechanistic knowledge of DNA replication and repair stress responses suitable to guide advanced intervention strategies for cancer. The conference will emphasize an environment suitable for initiating productive collaborations. It will include vibrant informal and formal discussions together with informative talks and poster sessions. To include late-breaking advances and innovating new investigators, one or more talks for every session will be selected from submitted abstracts. Meeting attendance is limited.

Keystone Symposia - Phosphoinositide Biology: New Therapeutic Targets Beyond Class I PI3K

Phosphoinositides have been recognized since the 1980s as important regulatory lipids supporting signal transduction from G protein-coupled receptors and receptor tyrosine kinases. The discovery of PI 3-kinases and their roles in carcinogenesis and immune regulation has created a fertile ground for translational expansion of the phosphoinositide field with a focus on cancer and inflammation. Most of these advances focused on Class I PI3Ks and PIP3-mediated signaling. However, research on other PI kinases and phosphoinositides has expanded substantially over the years. Class II and -III PI 3-kinases and less characterized 3-phosphorylated phosphoinositides are now rapidly emerging as key controllers of development, trafficking, nutrition-sensing and autophagy. Furthermore, phosphoinositides different from the 3-phosphorylated species are gaining momentum in many aspects of cell physiology and pathology. Exciting new developments include PI 4-kinases controlling vesicular trafficking and non-vesicular lipid transfer between membrane contacts, and new details are emerging on the role of inositol lipid phosphatases in trafficking, endocytosis and ciliary function. The recently recognized roles of PI 4-kinases as obligate host factors for certain RNA viruses, and their importance in parasitic organisms such as Plasmodium falciparum or Legionella pneumophila, have raised significant interest from pharmaceutical companies that now focus on inositol lipids other than the products of PI 3-kinases. By expanding our knowledge of phosphoinositides beyond class I PI3K products, this Keystone Symposia meeting represents a unique opportunity to bolster the rising interest in these under-explored avenues with a strong potential for translational impact.

A new community of researchers is emerging that focuses on studying and developing cellular therapies against cancer and chronic infections. The field is advancing rapidly due to our deepening understanding of cell biology. New technologies permit efficient cell culture, gene transfer and genetic editing, enabling autologous and allogeneic cell therapy. With rapid development of CAR T cells, breakthrough FDA approval status, and fast-tracking clinical trials, competition is fierce in the biotechnology and pharmaceutical industries. Cell therapies with stem cells and T cells are in advanced clinical development that will enable widespread use for cancer and chronic infections such as HIV. Infusions of regulatory T cells (Tregs) have the potential to benefit patients with autoimmunity and transplantation. A goal of this conference is to bring together basic and translational scientists from these fields to identify current opportunities and challenges in cell therapies. Another goal is to bridge the fields of effector T cell and Treg cell therapies. The interdisciplinary features of this meeting should facilitate new collaborations and interactions between investigators who would not normally meet. This meeting is paired with the Keystone Symposia conference on "Lymphocytes and their Roles in Cancer."

Keystone Symposia: Lymphocytes and their Roles in Cancer

Leverage of the body’s own immune system is revolutionizing the way that we treat cancer as it can lead to dramatic and durable responses. Such anti-tumor therapies include reactivation of anti-tumor T cells, expansion of tumor-infiltrating lymphocytes (TILs), Chimeric antigen receptor (CAR) T cells and a variety of cancer vaccines. The focus or target of most these approaches are cells of the lymphocytic lineage (T, B, NK, dendritic cells), key components of both adaptive and innate immune response. At the same time, lymphocytes can also be malignant, as in the case of a large number of lymphocytic leukemia and lymphoma subtypes, and active components of the immune microenvironment, controlling tumor growth and anti-tumor response. This conference for the first time brings together scientists from these different areas of lymphocyte biology and provides an overview of lymphocyte development, transformation, terminal differentiation, location within the tumor microenvironment and anti-tumor activity. The conference also provides an update on novel therapy protocols that attempt to harness the function of lymphocytes against cancer and state-of-the-art technologies, including imaging and DNA/RNA sequencing approaches, that will help us visualize and molecularly dissect anti-tumor responses in experimental systems and cancer patients.

In a constantly evolving oncology environment, ESMO continues to expand its educational offer and conference portfolio and is pleased to announce that as of 2018 ESMO is taking on the TAT Congress, which will now be held on an annual basis in Europe. The goals and the design of phase 1 studies have changed from just safety/toxicity to now include clinical activity/response and biomarker research. This new phase 1 paradigm has already led to rapid, conditional, approval of several new cancer therapeutics showing outstanding clinical activity in early-phase studies. Résumé:
The ECCO-AACR-EORTC-ESMO Workshop on Methods in Clinical Cancer Research is an educational programme that introduces junior clinical oncologists in any oncology subspecialty to the principles of good clinical trial design. In 2018, we will be celebrating 20 editions of this well-recognised and CME accredited Workshop. Over 1500 investigators have been trained at this Workshop, many of whom transformed into current world class experts pushing the boundaries of cancer research and therapy. One of the key benefits of the MCCR Workshop is exclusive access to and mentoring by up to 40 highly experienced clinical experts in the field of oncology from Europe, US and Canada. Under one roof, participants can engage with a multidisciplinary faculty, with expertise across the spectrum of cancers, to guide them through the complex and changing cancer research landscape to advance therapies which meet patients’ needs.

The conference coincides with the centenary of Stellenbosch University and forms part of our celebrations. In addition, the conference precedes the 18th International Congress of Endocrinology, which will be held at the Cape Town Convention Centre, Cape Town, South Africa, from 1-4 December 2018. The objective of this conference is to promote interaction and discussion within the field of steroid hormones and hormone-dependent cancers.
