Keystone Symposia - Aging, Inflammation and Immunity

The world’s aging population and problems with morbidity have been increasing rapidly. Decreases in immune function and increases in chronic inflammation with age are a major contributor to age-related diseases. This Keystone Symposia conference will provide a venue for researchers from around the world to discuss current findings and fundamental research to understand the mechanisms underlying declining immune function, increasing chronic inflammation, and possible interventions. The meeting will address essential aspects of aging/longevity research, with particular emphasis on the immune system and inflammation including effects of immunosenescence on cancer, autoimmunity, vaccine response and infectious disease. Longitudinal studies on aging and immunity, frailty and effects on mortality, quality of life and survival will also be covered as well as biomarkers of the aging immune system, mechanisms for generating chronic inflammation including the SASP, telomeres and the microbiome, regulation of aging and age-related immune function, and therapeutic approaches to slow aging and/or improve the immune system. The adaptive, innate and mucosal immune systems will be covered as these systems have a significant impact on human aging. Questions to be addressed include: 1) What changes in the immune system contribute to the observed decline in immune function and increase in the chronic inflammatory state? 2) How do diseases of aging or decreased immunity develop, and what can be done to prevent or reverse them? To address these questions, the symposium will gather investigators from interdisciplinary areas on aging, inflammation and immunology and will focus first on the human and secondly the murine immune systems. This meeting is paired with "Immunological Memory: Innate, Adaptive and Beyond."

In a constantly evolving oncology environment, ESMO continues to expand its educational offer and conference portfolio and is pleased to announce that as of 2018 ESMO is taking on the TAT Congress, which will now be held on an annual basis in Europe. The goals and the design of phase 1 studies have changed from just safety/toxicity to now include clinical activity/response and biomarker research. This new phase 1 paradigm has already led to rapid, conditional, approval of several new cancer therapeutics showing outstanding clinical activity in early-phase studies. Cambridge Healthtech Institute’s Third Annual Immuno-Oncology Summit Europe returns to the heart of London’s Canary Wharf for a full week of cutting-edge science and discussion. Through three back-to-back conference tracks, delegates have the opportunity to hear from the research teams at the forefront of the rapidly developing field of cancer biotherapeutics. The first track, Adoptive T-Cell Therapy, will highlight advances in next-generation CAR-Ts, TCRs, and TILs, and approaches for engagement of the innate immune system. The track will present genetic engineering technologies for T cells, preclinical models, and production, and focus on specificity, off-target toxicity and safety. A dedicated track on Combination Immunotherapy will follow with a review of strategies and approaches for designing rational combination immunotherapies. This track will provide case studies of immune modulator combinations, combinations of checkpoint inhibitors with other modalities, and biomarker-and mechanism-based selection of IO combinations. The final track of the week, Immunomodulatory Antibodies, will present advances with checkpoint inhibitors, agonistic receptor engagement, NK and macrophage activation, bispecifics, and Fc-engineering technologies. By 2025, most patients with cancer will receive immunotherapy as part of their treatment regimen. This symposium will cover the most critical topics integral to this vision: How can we integrate, sequence or combine immunotherapy with standard-of-care cytotoxic therapy, radiotherapy and targeted therapies? Which preclinical models are best predictive of combination approaches? How do we combine immune agonists as well as molecules inhibiting immune-suppressive pathways with checkpoint inhibitors? What are the rational combinations for cancer vaccines and viro-therapy? Which combinatorial approaches could prevent or treat adaptive mechanisms of resistance? Most immunotherapies are being developed in isolation, or being tested as single agents in preclinical models or in clinical studies. This conference helps to address the gaps in knowledge of how and when to combine therapies, and how to integrate immunotherapy into current standard-of-care or novel targeted therapy approaches – both preclinical and clinical. Overall, the objectives of this meeting are to: 1) Elucidate rational combinations for immunotherapy; 2) Discuss preclinical and other models to inform combinatorial approaches; 3) Understand the relevance for biomarker approaches; and 4) Provide insights into the latest preclinical and clinical data for immunotherapy combinations.

Keystone Symposia: Chromatin Architecture and Chromosome Organization

Understanding the principles governing genome regulation is one of the major challenges now facing biomedical research in the 21st century. Deciphering structure-activity relationships of the genome and its partners in the context of the cell nucleus is a necessary step for understanding the basis of development and disease, as well as for the elaboration of strategies against particular forms of cancers and genetic disorders. The genome is not a linear molecule of DNA randomly distributed in the nucleus, but exists as a three-dimensional (3D) object, intricately folded and packaged, structured around nuclear bodies and landmarks, acted upon by countless force-generating nano-machines and remodeling factors. With the recent advances that have been made in microscopy, biochemistry and modeling, the time is ripe to fully address the study of the genome in 3D space and time and consider it as a complex, dynamic biological system. Up until recently, many of the scientists involved in chromatin biology and epigenetics would attend meetings dedicated to this topic and would not necessarily interface with scientists using advanced imaging or physical and mathematical modeling approaches. This meeting will bring together some of the world’s leading experts and emerging talents at the interface of these topics to explore chromosome architecture and its dynamic relationship with genome function. Specifically, it will explore: 1) Chromatin structure and how this relates to gene expression and genome functions such as DNA replication and repair; 2) Chromosome conformation and the new insights into genome organization that have emerged using new technologies; 3) The nature of the chromatin partners (proteins, RNAs) that underlie chromatin folding and functions and their dynamic relationships in different contexts (cell cycle, development, environmentally induced changes, etc.); 4) Emerging imaging technologies and microscopy and the answers they are bringing to the dynamics of chromatin and chromosome architecture; and 5) New physical and mathematical modeling approaches to further our understanding of the principles governing chromatin architecture. Proper control of gene expression is important for all fundamental cellular and organismal processes. With advancements in technology and genetics, researchers are now able to use a combination of genetic, biochemical and computational methods to investigate gene control in healthy cells, as well as to discern what happens when these controls go awry in disease. Learning how gene expression programs are regulated is essential to understanding the control of cell state and its impact on disease development. Multiple levels of gene regulation including basic transcriptional controls, enhancers, epigenetics and the role of phase separation in transcriptional control will be addressed. The goal of this conference is to share new insights into gene control in health and disease and to discuss advances that are being made in disease therapy based on gene control therapeutics. The 2018 Gordon Research Conference on DNA Damage, Mutation and Cancer is developed around the theme of exploiting fundamental knowledge to advance treatment and prevention. Rich and ever-increasing information is available on the specific mutations that are the molecular basis of cancer. Many mutations can be traced to specific types of damage to the DNA, while others arise from DNA replication errors or unknown causes. Technologies are being refined to determine the origins of these mutations, and monitor the accuracy and reproducibility of cancer genomic data. Biological and structural analyses can assess which mutations are functionally relevant. Specific mutations can confer vulnerabilities, making individual cancers more responsive to treatment with DNA damaging agents.

We are living in very exciting times in the context of molecular oncology. A deeper understanding of the molecular mechanisms involved cancer is revealing novel therapeutic approaches to treat cancer. A new and exciting therapeutic target has emerged recently. LIF is a cytokine aberrantly expressed in cancer that acts as a pleiotropic oncogenic factor. It regulates cancer stem cells, the tumour microenvironment and specifically the immune system. In this conference, we will learn about the most recent advances in the field of LIF from the basic science to clinical studies including the understanding of its biology, its implications in cancer and the development of anti-cancer agents. The target audience for this conference is basic and translational scientists and clinicians, both senior and young investigators that want to have a broad and detailed overview of the most recent advances in the field of LIF.

The ECCO-AACR-EORTC-ESMO Workshop on Methods in Clinical Cancer Research is an educational programme that introduces junior clinical oncologists in any oncology subspecialty to the principles of good clinical trial design. Over 1500 investigators have been trained at this Workshop, many of whom transformed into current world class experts pushing the boundaries of cancer research and therapy. One of the key benefits of the MCCR Workshop is exclusive access to and mentoring by up to 40 highly experienced clinical experts in the field of oncology from Europe, US and Canada. The Association's top management selected Russia as the venue for the 30th Congress. This historical event for the Russian medical community will be held in Moscow on September 9-12, 2018. Burnazyan Federal Medical and Biophysical Center at FMBA of Russia, IASGO's official representative in Russia, is the lead arranger of the Congress.
