Bacterial infections represent an increasingly urgent threat to human health worldwide with major pathogens being resistant to all available antibiotics. In addition, antibiotic therapy can fail to clear bacterial infections even in the absence of clinically detectable resistance. This is likely caused by drug-tolerant bacteria that survive even extensive exposure to antibiotics. The investigation of underlying mechanisms of microbial persistence and antibiotic tolerance is a newly emerging research field with already spectacular and fascinating insights. The “Bacterial Persistence and Antimicrobial Therapy” Conference will bring together the world’s leading experts in this field using a variety of approaches.

FASEB SRC: Biological Methylation: Fundamental Mechanisms in Human Health and Disease

This FASEB SRC focuses on recent advances in our understanding of the role of dynamic methylation of proteins, DNA, and RNA in diverse nuclear and cytoplasmic biological processes and how this modification impacts epigenetic cellular identity, tissue homeostasis, development, and the pathogenesis of human diseases. Since methylation has become a major therapeutic target in recent years, the development of epigenetic-based medicines that target methyltransferase, demethylase, and methylation readers for the treatment of cancer and other diseases will be a topic covered in the program.

FASEB SRC: Functional Disulfide Bonds in Health and Disease

Most proteins are chemically modified after they are made to control how, when, and where they function. It was thought that only the amino acid side chains and the peptide bonds of proteins were chemically modified. It has become clear in recent years that the disulfide bonds of proteins are also modified. Some disulfide bonds are cleaved in the mature protein to control function. From a few examples a decade ago, there are now about thirty well-defined examples of control of protein function by functional disulfides and dozens more examples that are under investigation. Cleavage of a functional disulfide in a protein changes the bioactivity of the protein. Changes in ligand binding, substrate hydrolysis, proteolysis, or oligomer formation have been described. The functional disulfides are cleaved by reductases or by thiol–disulfide exchange. Viral, bacterial, plant and mammalian proteins are regulated by functional disulfides, so this mechanism of protein control operates in all life forms. The early indications are that cleavage of disulfide bonds may rival proteolysis of peptide bonds as a means of protein control.

This FASEB SRC will provide focus on the diverse topics such as: Cell Surface Exposed Glycans of Bacteria and Archaea, host-pathogen interactions, Microbial Symbiosis and Dysbiosis, polysaccharide-based vaccines and therapeutics, industrial applications including biofilms and glycoengineering. The common element of glycosciences in this diverse group fuels the collaborative projects and the cross-fertilization of ideas that make this field exciting for many researchers in academia and industry. The organizers and chairs will invite persons to speak on a wide variety of subject matter (model systems, diseases, macromolecules, etc) and technologies (genetics, mass spectroscopy, X-ray, NMR, etc ). FASEB SRC: Translational Neuroimmunology

This SRC is focused on defining the contributions of neuroimmune interactions in CNS health and disease and explores the opportunities and challenges for harnessing neuroimmune interactions as therapies for classic neurologic and neurodegenerative diseases including neurodevelopmental disorders, TBI, spinal cord injury, stroke, epilepsy, multiple sclerosis, Alzheimer’s and Parkinson’s disease. This conference explores recent cutting edge discoveries on the molecular mechanisms of neuro-immune interactions involved in maintaining or disrupting CNS health as well as the influences of the microbiome, environmental toxicants and genotype on tipping the balance between health and disease. Aspects of this SRC conference that have been ranked highly by past attendees include, ample presentation and discussion time for oral presentations, selection of abstracts from junior scientists and graduate trainees for short platform presentations, well attended poster sessions, time for free and open discussions between speakers and attendees aimed at fostering new and ongoing collaborations. New this year are addition of two workshops discussing pragmatic issues and careers aimed at bridging the well known “valley of death” between seminal research discoveries and development of therapies. Altogether, attendees of this SRC will enjoy a multidisciplinary group of researchers not usually found together at specialized conferences and will benefit from the focus on the opportunities and challenges for translating basic research discoveries to therapies.

FASEB SRC: The Lung Epithelium in Health and Disease

This FASEB SRC will bring together scientists in academia and industry for live discussions on the most recent progresses in the field of lung epithelium biology. The epithelium of the lung allows airway flow and provides an essential barrier against infection and other detrimental exposures, which is made possible through highly orchestrated processes during development, homeostasis and repair. The goal of this meeting is to advance our understanding of these fundamental processes by providing a highly interactive platform for senior and junior scientists with expertises in biology, disease models and techniques. Topics of special interest in this FASEB meeting include airway development, stem cells and regeneration, cell-cell interactions that orchestrate injury responses and repair, and the impact of newest tools on lung biology research. Together, we believe we will gain insights into mechanisms of airway diseases that involve single gene, complex genetic interactions, and environmental impact. The knowledge will lay the foundation for the identification of new drug targets and the improvement of treatment strategies for devastating lung diseases, such as asthma, COPD, and cystic fibrosis.

Keystone Symposia: Framing the Response to Emerging Virus Infections

Résumé:
Emerging and re-emerging viruses have the potential to cause high morbidity and mortality and range from localized outbreaks to epidemics. Due to their emerging nature, most aspects of the biology and infectious potential of these viruses are poorly understood. Our continuing struggle to respond to a procession of pandemics including SARS, avian influenza, MERS, Ebola and more recently Zika, highlights key gaps in our knowledge and should serve to motivate our re-thinking on how we can better prepare and deal with future unknown viral threats. This meeting will focus on important areas such as surveillance, diagnostics and countermeasures and other important advances in new technologies and how they are being applied to research. Furthermore, we will discuss how to facilitate the translation of research, data and candidate treatments through the development pipeline in a timely and cost-effective manner. The key themes to be covered include the need to understand why zoonotic diseases matter, their association with agriculture, the importance of surveillance and early detection, and the difficulties of dealing with diseases that involve both medical and veterinary communities. The conference will bring together experts in virology, immunology, vaccinology, epidemiology with those that seek to transfer knowledge between these groups, veterinarians and industry and government. Further, this meeting would bring together individuals involved in the control of these diseases in government and non-government organizations, as well as people involved in study of zoonosis and countermeasures. The creation of global networks and sharing of information will ensure that we are better prepared for future outbreaks.

The programme is of interest to clinical investigators from across the globe to review current research and discuss its implications for HIV management strategies. Although there has been steady progress in HIV drug treatment, significant challenges still remain, including persistence of elevated risk of non-AIDS morbidity, with potential for HIV and/or antiretroviral therapy (ART) as causes, paediatric and adolescent care, models of care for the ageing population, and treatment issues throughout less well-resourced parts of Europe and the rest of the world. The Congress is also moving into areas beyond treatment, such as prevention, with time in the programme focussed on pre-exposure prophylaxis (PrEP) and issues across Europe and beyond. The important role of people living with HIV in treatment decisions and self-management continues to feature, and we look forward to a number of perspectives in the programme from the community.

Bacteria and fungi residing in the female genital tract have been associated with elevated risks of HIV acquisition and transmission, pelvic inflammatory disease, miscarriage, preterm birth, and invasive maternal and neonatal infections. The increased availability of high throughput genomic testing since the turn of the century has revealed a more detailed picture of these organisms than was possible when evaluation depended on microscopy and culture. The interrelationships between sexually transmitted infections, vaginal dysbiosis, vulvovaginal candidiasis, and vaginal pathobiont carriage are being elucidated, and their effects on the cervicovaginal mucosal barrier and immune system are being characterized. The mechanisms that may lead to adverse outcomes are being unraveled, and an increasing number of interventions are in clinical trials. In this meeting, the current understanding of female and male genital tract microbiology and immunology, including functional microbiology and biofilms, will be presented and knowledge gaps identified. Potential mechanisms leading to adverse outcomes, and a variety of potential prevention and/or treatment interventions, will be presented and discussed. The exponential progress made in recent years will hopefully lead to efficacious public health interventions to reduce the high prevalence of adverse sexual and reproductive health outcomes in women, especially in resource-poor settings.
