CRISPR/Cas — CRISPR/Cas9 genome editing in model and non-model organisms – from theory to the bench

Zusammenfassung:
The Department of Life Sciences at Shiv Nadar University is organizing a 5-day residential, practical training workshop titled "CRISPR/Cas9 genome editing in model and non-model organisms from theory to the bench” from 24th to 28th May, 2018. The workshop will provide hands-on training in genome editing of mammalian cells as well as of infectious pathogens using the state-of-the-art CRISPR/Cas9 system. Participants will learn to develop a CRISPR/Cas project suitable to answering their research question, identify and design optimal CRISPR targets using bioinformatics tools, generation of gene knockouts, and validation of knockouts using the most current technologies. Quantitative Perspectives on Cancer

Zusammenfassung:
From a clinical point of view, cancer is a devastating disease. From the point of view of theoretical physics, it is fascinating. Cancer progression involves cells changing shape, cells that migrate under constraints imposed by the human body, flow of blood and cells, and changing of the physical characteristics of a human tissue, to give just a few examples. Moreover, some aspects of cancer development, such as population genetics, are well rooted within subfields of theoretical physics. In this conference, we will host world-renown experts in cancer research who will present their views of fundamental problems that limit our understanding of cancer (and where theoretical physics might be helpful). In addition, leading physicists and theoretically-oriented scientists from related fields will discuss theory and modelling applied to cancer research, and the presence of cancer biologists will facilitate a dialogue between the disciplines

Oscillations are abundant - from hormonal oscillations with periods of days and months, to genetic fluctuations in the range of hours (circadian clock, embryonic oscillators), and metabolic and biochemical oscillations in the order of minutes (glycolytic oscillations) and seconds (Calcium-dynamics) - oscillations are central to biology across different temporal and spatial scales. At this symposium multiple disciplines will come together in order to discuss common mechanisms underlying the generation of oscillations.This symposium aims to further strengthen a new, highly interdisciplinary community composed of scientists from very different fields and who share a common interest in oscillatory phenomena and biological dynamics. As such, this meeting provides a unique platform for this emerging new community working at the interface between quantitative biology, complex oscillatory systems and physics.

Utilising Flow Cytometry we can identify and quantify different from heterogeneous samples, analyse the corresponding state of proliferation or simply detect the expression of fluorescent reporters. Improvements in assay technology are now enabling scientist to detect mRNAs in cells using flow cytometry. We will cover the theory of Flow Cytometry and corroborate this information intensively with the practical issues of the most frequent assays as part of our practical course curriculum.

2018 Ross Prize in Molecular Medicine — Genetics of Neurodevelopmental Disorders

The 2018 Ross Prize in Molecular Medicine will be awarded to Dr. Huda Y. Zoghbi for her research unveiling the genetic and molecular basis of Rett syndrome and spinocerebellar ataxia, enabling novel therapeutic strategies for these devastating diseases. Her discovery of the mutational driver of Rett syndrome also lead to the elucidation of its role in other neurological impairments, ranging from learning disabilities to autism. Complemented by fundamental studies in neurodevelopment, Dr. Zoghbi continues to pursue the complex molecular processes driving the pathogenesis of some of the most devastating neurological conditions.

The Ross Prize in Molecular Medicine was established in conjunction with the Feinstein Institute for Medical Research and Molecular Medicine to recognize biomedical scientists whose discoveries transformed the way medicine is practiced. The awardees are midcareer researchers who have made a significant impact in the understanding of human disease pathogenesis and/or treatment. Moreover, it is anticipated that they will continue to make profound advances in the general field of molecular medicine.

The course participants will use open-source software. R-coding skills and deep knowledge of statistics are not necessary for attendance because most analyses will be performed with docker4seq package, which was developed to facilitate the use of computing demanding applications in the field of NGS data analysis. Docker4seq package uses docker containers that embed demanding computing tasks (e.g. short reads mapping) into isolated containers. This approach provides multiple advantages:        •     user does not need to install all the software on its local server; •     results generated by different containers can be organized in pipelines; •     reproducible research is guarantee by the possibility of sharing the docker images used for the analysis. Computers for hands-on exercises will be provided along with demo data sets.

The 2018 Gordon Research Conference on Cyclic-Nucleotide Phosphodiesterases (PDEs) will focus on efforts to develop the therapeutic potential of phosphodiesterase modulators while also highlighting new biology relevant to the PDEs. As the sole means for terminating the intracellular signals carried by ubiquitous second messengers cAMP and cGMP, the PDEs are uniquely positioned to regulate a host of signaling cascades involved in cell to cell communication, metabolism and gene regulation. Unique elements of the quaternary structure of each family has allowed for the generation of selective inhibitors despite the high level of sequence identity in the active site of the enzymes. Only two families, PDE 6 and 11, still await the development of useful preclinical tools/inhibitors and drug candidates are in or nearing human studies for PDE 1, 2,3,4,5, 9 and 10. Clinical data is accumulating on the effects of PDE 3, 4, 5, 9 and 10 inhibition although only the effects of PDE5 inhibition have been subject to extensive, albeit poorly undocumented, profiling.

This FASEB SRC will present new discoveries in Ca2+ signaling at the basic and translational levels. Calcium is the most ubiquitous second messenger in biology and plays critical roles in a wide range of dynamic process from synaptic transmission to sensory perception, development, gene expression, cell migration, the immune response, and heart function. Defects in Ca2+ signaling lead to a correspondingly large number of serious diseases, including nervous disorders (stroke, mental illness, neurodegenerative diseases), immunodeficiencies, hypertension, cardiac arrhythmias, and developmental defects.

This FASEB SRC will bring together international scientists from different backgrounds to discuss cutting-edge developments in the areas of immunoreceptors and immunotherapy. Immunoreceptors encompass an array of receptor types, including B/T cell receptors, cytokine and immunomodulatory receptors that play a central role in cancer, autoimmunity and infectious disease. Although the field of immunoreceptors is well established, there are significant growth areas that continue to drive the field forward. This FASEB SRC is the next iteration of a liver-focused meeting that has been held biennially since 1988. It is recognized as the premier small conference covering all aspects of basic research involving the liver. The main goals of the conference are to i) present and integrate research advances in disparate areas of liver biology in order to foster new national and international collaborative projects, ii) encourage junior and early stage investigators, women and minority scientists to pursue liver research to ensure that the study of hepatic disease and biology remains vigorous and iii) accelerate translation of fundamental advances into novel clinical treatments through the interactions of basic and clinical researchers. These goals will be achieved through specific programmatic strategies.

The program will bring together a group of theoretical physicists, mathematicians, experimental biologists, and experimental biophysicists, to study the mechanisms by which (i) protein assemblies, such as biopolymers, exert forces in cells, (ii) protein assembly dynamics in cells are regulated by forces, and (iii) the dynamics of force-generating assemblies are controlled by signaling pathways. The program will enhance the formation of collaborative links between experiment and theory, as well as those between theorists using different methodologies. It will also clarify the links between apparently disparate, but related phenomena by bringing in individuals with a broad range of backgrounds. Finally, it will enhance the development of the mechanobiology community in the Nordic countries.

This FASEB SRC will focus on membranes and membrane proteins. Membranes define the boundaries of cells and sub-cellular compartments. Membrane proteins, which represent over 25% of proteins and over 50% of drug targets, play key roles in many life processes. Advances in methodology have led to tremendous recent progress in this field. For instance, dramatic increases in the number of membrane protein structures known at atomic resolution provide the foundation for mechanistic studies and drug development efforts. Advances in microscopy methods are enabling visualization of biological processes involving membranes, and structure determination of large membrane-bound complexes. The conference program includes sessions on a variety of membrane proteins and complexes, as well as on the complexities of the membrane itself – the roles of its domains and shapes in various biological processes. Topics will include mechanisms and folding of membrane proteins of both fundamental and medical importance.

FASEB SRC: Functional Disulfide Bonds in Health and Disease

Most proteins are chemically modified after they are made to control how, when, and where they function. It was thought that only the amino acid side chains and the peptide bonds of proteins were chemically modified. It has become clear in recent years that the disulfide bonds of proteins are also modified. Some disulfide bonds are cleaved in the mature protein to control function. From a few examples a decade ago, there are now about thirty well-defined examples of control of protein function by functional disulfides and dozens more examples that are under investigation. Cleavage of a functional disulfide in a protein changes the bioactivity of the protein. Changes in ligand binding, substrate hydrolysis, proteolysis, or oligomer formation have been described. The functional disulfides are cleaved by reductases or by thiol–disulfide exchange. Viral, bacterial, plant and mammalian proteins are regulated by functional disulfides, so this mechanism of protein control operates in all life forms. The early indications are that cleavage of disulfide bonds may rival proteolysis of peptide bonds as a means of protein control.

This FASEB Science Research Conference, the 14th in the series, showcases the most recent advances in our understanding of neuronal circuits responsible for retinal image processing. With its loyal following of accomplished and passionate scientists, it has evolved into one of the premier international meetings on retinal circuitry, development, and information coding. A broad range of concepts and approaches will be covered in platform talks featuring internationally recognized leaders in the field as well as junior faculty and advanced postdoctoral trainees. These will be grouped into thematic sessions, with topics including retinal direction selectivity; information processing at photoreceptor and bipolar-cell synapses; intrinsically photosensitive ganglion cells; retinal development; the structure and function of specific types of bipolar, amacrine and ganglion cells; and translational visual neurobiology, including stem cells and other regenerative approaches. This FASEB SRC will unite scientists pushing the boundaries of our molecular understanding of the nucleic acid transactions which facilitate life. Information encoded in RNA and DNA is accessed, maintained, decoded and degraded by a staggering number of proteins, protein/DNA and protein/RNA complexes that catalyze specific fundamental enzymatic reactions which drive nucleic acid transactions— not limited to, but including key cellular processes such as DNA replication and repair, recombination, transposition, transcription, translation and epigenetic gene regulation. These protein and RNA machines are impacted in varying disease states, and can be exploited for therapeutic advantage including using CRIPSR gene editing technologies. The use of emerging biophysical methodologies such as single-molecule spectroscopy/microscopy and small-angle X-ray scattering, cryo electron microscopy, and chemical biology has opened new doors to exploration of nucleic acid metabolism. This meeting will highlight exciting efforts underway to decipher the anatomy, architecture and mechanisms of these protein and protein-nucleic acid machines in order to understand how they recognize, engage, and manipulate the structure of DNA and RNA. We aim to unite established and future leaders in the field to exchange ideas and formulate new collaborations. We highly encourage student, postdoctoral trainees, and young investigators to attend, and will encourage their attendance with student and postdoc travel scholarships and a large number of oral presentations, to be selected from the abstracts. Selected talks, poster presentations, and recreational activities will present students and postdoctoral fellows with both formal and informal networking opportunities. Additional organized "meet the experts" panel discussions will help junior colleagues navigate various topics related to careers and women in science. This exciting program will facilitate cross-talk between multiple areas of science, techniques, and skill levels to inspire new innovations and quality science in the area of nucleic acid structural and mechanistic biology.

FASEB SRC: Dynamic DNA Structures in Biology

For decades after its discovery, genomic DNA was believed to exist only as a right-handed, double helix known as B-DNA. Its sole functions were to serve as a template for RNA synthesis or as a template for the production of identical duplexes during chromosome replication. These core beliefs were shaken by the discovery that DNA is covalently modified and packaged into chromatin, and that the molecule can form an enormous variety of non-B alternative structures, including cruciforms, a left-handed helix, three- and four-stranded helices, and slip-strand configurations. Transient denaturation of the duplex during all major DNA transactions (replication, transcription, repair and recombination) promotes dynamic transitions to non-B DNA structures as well as other structures of interest such as R-loops, D-loops, reversed replication forks, Holiday junctions, and other complex DNA-DNA or DNA-RNA structures. Furthermore, repetitive DNA sequences, which are highly overrepresented in genomic DNA, are particularly prone to structural transitions. Studies conducted in many labs worldwide have confirmed that structural transitions are not only central to normal functions of the genome, but also are responsible for occasional malfunctioning that leads to a disease state. The most striking example is expansion of structure-prone DNA repeats, which is responsible for more than thirty hereditary neurological and developmental diseases.

This SRC is focused on defining the contributions of neuroimmune interactions in CNS health and disease and explores the opportunities and challenges for harnessing neuroimmune interactions as therapies for classic neurologic and neurodegenerative diseases including neurodevelopmental disorders, TBI, spinal cord injury, stroke, epilepsy, multiple sclerosis, Alzheimer’s and Parkinson’s disease. This conference explores recent cutting edge discoveries on the molecular mechanisms of neuro-immune interactions involved in maintaining or disrupting CNS health as well as the influences of the microbiome, environmental toxicants and genotype on tipping the balance between health and disease. Aspects of this SRC conference that have been ranked highly by past attendees include, ample presentation and discussion time for oral presentations, selection of abstracts from junior scientists and graduate trainees for short platform presentations, well attended poster sessions, time for free and open discussions between speakers and attendees aimed at fostering new and ongoing collaborations. New this year are addition of two workshops discussing pragmatic issues and careers aimed at bridging the well known “valley of death” between seminal research discoveries and development of therapies. Altogether, attendees of this SRC will enjoy a multidisciplinary group of researchers not usually found together at specialized conferences and will benefit from the focus on the opportunities and challenges for translating basic research discoveries to therapies.

Microfluidics is a quickly growing, highly interdisciplinary field at the interface of physics, engineering, chemistry and biology. Over the past years microfluidic approaches have been used for a variety of applications, including nucleotide sequencing, functional genomics, single cell/ single molecule studies and diagnostics. Many of these applications, including next-generation sequencing devices, have been revolutionised by miniaturisation, paving the way for global gene analysis and hence transforming biology. In addition, working on the pico- to nanoliter scale not only allows a high degree of parallelisation, but also offers superb spatiotemporal control. Small objects such as cells, or even discrete parts thereof, can be exposed to unique conditions, facilitating entirely novel approaches in modern biology and chemistry.

This FASEB SRC focuses on protein phosphorylation - the most prevalent post-translational modification that regulates almost all aspects of development and normal physiological function of an adult organism. Decades of research has revealed the complexity of the regulatory networks involved in control of protein phosphorylation and their exceptional vulnerability as target of dysregulation in many disease states. Protein kinases, which catalyze phosphorylation, are one of the most successful targets of currently FDA-approved therapeutics, but emerging mechanisms of drug resistance mandate deeper mechanistic understanding of their function, as well as investigation of innovative alternative strategies to targeting the aberrant phosphorylation networks.

This FASEB SRC will focus on virus assembly, virus structure, and structure-based mechanisms of virus replication (entry, replication, virus-host interactions). Studies in these areas provide insight into a multitude of cellular processes from the perspective of relatively simple and relatively easy to manipulate organisms, viruses and bacteriophages. Moreover, these cellular processes are critical for the pathogenicity of infectious viruses and therefore are targets for established and novel antiviral therapeutics. Many new therapies are a direct result of basic scientific investigations, which have been the emphasis of past FASEB Virus Structure and Assembly conferences.

This FASEB SRC addresses the process of how proteins acquire their beautiful three-dimensional structures, required for functionality. Protein folding has been studied for decades in the test tube, and while this approach continues to aid our understanding of the fundamental physical and chemical forces that govern folding, there has been a concomitant and growing appreciation that the cellular environment presents unique challenges – as well as unique support systems – that cannot be easily captured in the test tube. Understanding how proteins fold and why they misfold are major goals in the quest to understand the mechanisms of the cellular protein quality control machineries, The knowledge gained will help us to find strategies to correct misfolding diseases, which include Alzheimer’s and cystic fibrosis.

One-carbon metabolism and related dietary factors (folic acid and vitamin B12, as well as vitamin B6, choline, riboflavin, methionine, serine, cysteine, and others) play important roles in a vast array of essential physiological functions. Disruptions of one-carbon metabolism due to dietary, pharmacological, environmental, and genetic factors impact most of the major health issues of our time, including vascular disease, cancer, neurodegenerative disease (Alzheimer’s disease and dementia, Parkinson’s disease), birth defects, and comorbidities associated with obesity and diabetes such as renal disease. The “Folic Acid, Vitamin B12, and One-Carbon Metabolism” SRC has been meeting every other year since the mid-1980s. From its inception, the conference has embraced an integrative philosophy that brings together basic researchers, physician scientists, and policy makers to share cutting edge research and to discuss its ramifications for health at both the personal and population levels. For 2018, in addition to updates on new findings related to the fundamental biochemistry of folates, B12, and one-carbon metabolism, session topics will focus on 1) Vitamin B12 transporters: binders, receptors & chaperones, 2) Oxidative stress, one-carbon metabolism and development, 3) In utero effects of disruption in one-carbon metabolism, 4) One-Carbon metabolism and neurodegenerative disease, 5) Choline – The good, the bad and the ugly, 6) Interactions between pharmacologic agents, xenobiotics and 1C metabolism, 7) Novel aspects of transsulfuration, and 8) The many faces of MTHFR. Conference attendees can expect thought-provoking oral communications from leaders in the field, stimulating discussions during questions and answers sessions and expanded opportunities for students and post-doctoral trainees to present their work in both oral and poster sessions, as well as formal and informal opportunities to exchange ideas and establish new collaborations.

This FASEB SRC will bring together scientists in academia and industry for live discussions on the most recent progresses in the field of lung epithelium biology. The epithelium of the lung allows airway flow and provides an essential barrier against infection and other detrimental exposures, which is made possible through highly orchestrated processes during development, homeostasis and repair. The goal of this meeting is to advance our understanding of these fundamental processes by providing a highly interactive platform for senior and junior scientists with expertises in biology, disease models and techniques. Topics of special interest in this FASEB meeting include airway development, stem cells and regeneration, cell-cell interactions that orchestrate injury responses and repair, and the impact of newest tools on lung biology research. Together, we believe we will gain insights into mechanisms of airway diseases that involve single gene, complex genetic interactions, and environmental impact. The knowledge will lay the foundation for the identification of new drug targets and the improvement of treatment strategies for devastating lung diseases, such as asthma, COPD, and cystic fibrosis.

This FASEB SRC focuses on the role of the immune system in the development and exacerbation of the many conditions that encompass metabolic syndrome. Obesity and metabolic syndrome have increased at epidemic proportions over the past two decades. Our understanding of how the immune system is influenced by adipose tissue has evolved greatly, even as this epidemic continues to rage. Once considered as primarily a fuel storage organ, adipose tissue is now known to have a number of immunomodulatory properties, which affect whole body physiology, and importantly, are thought to account for many of the complications of obesity including insulin resistance and cardiovascular disease. The presence and activity of immune cell types such as macrophages and T cells within adipose tissue is more recently being teased out given the appreciation of obesity and insulin resistance as inflammatory in nature. It is becoming increasingly clear that in addition to altering adipose tissue function, immune cells impact the physiology and homeostasis of other organs such as the liver, CNS, skin and intestine in obesity. The immunological and inflammatory influences of obesity contribute to the complications that make up metabolic syndrome including insulin resistance, diabetes, and cardiovascular disease.

We are pleased to announce the 3rd HVP Variant Effect Prediction Training Course at Newcastle University Medicine Malaysia, (only 25 Km to Singapore). On top of the valuable insights shared by our distinguished speakers, discussions and workshops will also be organised for delegates to interact and engage with hands-on practical workshops. The course will be a mix of theory and practice presented by invited expert speakers and abstract-selected participant contributions. Practical sessions will be a mix of academic and commercial demonstrations giving course participants hands-on experience with computational analysis tools. The course is tailor-made for people working in a DNA laboratory including clinical diagnostic labs involved in DNA sequencing data analysis. Participants are Expected to bring their own laptop.

Solving structures of large macromolecular assemblies to gain fundamental insights into biological mechanisms remains a major challenge in structural biology. Cryo-EM combined with image processing is currently undergoing what has been referred to as a “resolution revolution”, making it one of the most powerful techniques to address this challenge. It also has unique capabilities to obtain structural information in situ within complex environments, and to deal with heterogeneous samples. The methods are complex and under constant development, so in depth, advanced training of new users is essential to ensure future users can adapt new technologies to expand into new areas and solve new biological problems. This course provides the theoretical and practical training covering the full breadth of cryo-electron microscopy (cryo-EM) techniques from sample preparation to data collection to image processing.

Chromosomes are arguabely one of the most important structures inside cells. Several meters of DNA are compacted into micrometer-sized assemblies in every human cell. Recent advances in our understanding of how this is achieved, and of its implications on nuclear functions, have been breathtaking. Instead of black boxes, or imaginary text-book style folding models, advanced imaging and genomics techniques have layed the foundation for the first well-informed molecular models of chromsomes. These portray chromosomes as largley self-organising entities of nucleosome-melts, organised by the universal SMC complexes and additional chromosomal proteins. Recent work culminated in the biochemical reconstitution of chromosome formation from defined components (Hirano).

he meeting will cover structural/mechanistic studies of processes using super-resolution imaging and single-molecule methods (tracking, bleaching, co-localisation, FRET). Many processes involve nucleic acids (chromosome organization, DNA replication, repair, recombination, and segregation, gene expression, CRISPR mechanisms), whereas others relate to signal transduction, cell division, cell-wall biosynthesis, and motility. The meeting will also explore new quantitative in vivo techniques applicable to bacteria: theoretical approaches (modelling stochastic gene expression), microfluidics, site-specific labelling, cytological profiling, high-throughput approaches). A third theme will involve studies of clinical/commercial significance (biofilms, pathogenic bacteria, bacterial persistence, antibiotic resistance).

EMBL Course: Attacking Open Chromatin with ATAC Sequencing

ATAC-Seq stands for Assay of Transposase Accessible Chromatin and is, despite its recent invention, already a widespread method to profile open chromatin on a genome-scale. The goal of this course is to teach the entire workflow of an ATAC-Seq experiment including library preparation, sequencing, data processing, and downstream analysis by addressing a real biological question that will be answered at the end of the course. We will start with the experimental ATAC-Seq protocol, highlighting the critical steps, potential pitfalls, and the most up-to-date tweaks to improve the quality of the results. After sequencing the samples we will go through the basic quality control and preprocessing steps using multiple analysis tools for next generation sequencing data. Finally, we will perform different types of analysis on the processed data, including visualisation, differential peak calling, and differential transcription factor activity profiling to come to a biological conclusion. In addition we will discuss recent advancements in technology development and algorithms for analysis and give an outlook on future perspectives and applications.

The fields of membrane trafficking in general and autophagy in particular got the Nobel Prize for Medicine in 2013 and 2016, respectively. This work laid the foundation for understanding these processes at a mechanistic detail and high precision. The question of how membrane structure and function is regulated is highly relevant to human health and disease. Regulation of membrane-associated processes is a very dynamic field relevant to various disciplines, from neuroscience to cell and developmental biology. Small GTPases have emerged as important regulators of all membrane trafficking processes. These GTPases together with their various interactors regulate intracellular trafficking, organelle dynamics, proteostasis, signaling and metabolism. Mutations in these molecules result in human diseases, from inherited to complex disorders. For example, dis-regulation of small GTPase networks has been implicated in multiple diseases, including cancer and neurological disorders.

The goal of this course is to teach the fundamentals of NGS library preparation and validation using the Illumina TruSeq stranded total RNA sample preparation kit. We will start with an introduction into Illumina NGS technology and the library preparation workflow. The main part will be the preparation of libraries from total RNA, with a focus on the critical steps and potential pitfalls of the protocol. Finally we will quantify and quality control the prepared libraries, before discussing solutions for total RNA sequencing data analysis.

This SRC will focus on fibroproliferative diseases, including pulmonary fibrosis, systemic sclerosis (skin), liver cirrhosis, cardiovascular disease, progressive kidney disease, and macular degeneration. Fibrosis can affect all tissues and organ systems. Fibrotic tissue remodeling can also influence cancer metastasis and accelerate chronic graft rejection in transplant recipients. Nevertheless, despite its enormous impact on human health, there are currently no approved treatments that directly target the mechanisms of fibrosis. A better understanding of the disease mechanism of fibrosis across organs is of high interest and importance to the scientific community.

Keystone Symposia: Drivers of Type 2 Diabetes - From Genes to Environment

The prevalence of diabetes mellitus is increasing worldwide and is a major threat to global public health that requires urgent action. Over the last few decades, significant advances have been made in terms of identifying novel susceptibility genes and signaling pathways that play pivotal roles in the pathogenesis of diabetes and its related metabolic disorders. However, a major gap in understanding the current global epidemic of diabetes is the lack of knowledge regarding how molecular interactions between the environment and the susceptibility genes are regulated within an altered metabolic milieu. This meeting will address these unresolved gaps in the etiopathogenesis of diabetes and focus on the latest advances that are linked to the molecular drivers of diabetes mellitus. Topics include: systemic regulation of adipocytes in diabetes; microvesicles, noncoding RNA, and intercellular communications; physiological drivers in hunger and energy homeostasis; epigenetics and metabolic control in diabetes; novel signaling players related to insulin resistance; adaptation of beta cell to chronic metabolic stress; environmental triggers in diabetes and metabolic diseases; and molecular targets for nutrient sensing and signaling. The organizers anticipate that this meeting will bring about a major shift in addressing the causes of diabetes mellitus, as the topics emphasized in this meeting have not yet been explored widely. The course participants will learn how to use R/BioConductor software and Illumina BaseSpace Cloud apps. The course covers various topics using graphical interfaces. Furthermore everyday there will be, in morning and afternoon, a brief session on R scripting to provide the basics knowledge to move from the use of the graphical environment to command line. Computation is an integral part of today's research as data has grown too large or too complex to be analysed by hand. An ever growing fraction of science is performed computationally and many wet-lab biologists spend part of their time on the computer. Many scientists struggle with this aspect of research as they have not been properly trained in the necessary set of skills. The result is that too much time is spent using inefficient tools when progress could be faster. This is a course for researchers in the life sciences who are using computers for their analyses, even if not full time. The target student will know a little bit of command line/programmatic computer usage, but not consider themselves an expert. A target student will have written a for loop in some language before, but will not know what git is (or at least not be very comfortable with advanced git usage).

Systems genetics is the extension of genetics to the system-level. It aims at understanding the relationship between genotypes and molecular phenotpes at the genome-scale, thus integrating genomics with large-scale molecular and phenotypic profiling. This opens the door to the investigating of the genetic basis of complex traits in a variety of model systems, from yeast growth phenotype to human disease. Taking advantage of natural genetic variation allows to investigate the effect of many mild genetic perturbations, an approach which contrasts with the all-or-none targeted genetic perturbations.

Keystone Symposia: From Rare to Care - Discovery, Modeling and Translation of Rare Diseases

Work over the past 30 years has resulted in the identification of genes for ~50% of the estimated 7,000 rare genetic diseases; it is predicted that most of the remaining disease genes will be identified in the next 10 years. Approximately 500 medicinal products are currently on the market for rare diseases. The accelerating pace of rare disease gene identification means, in effect, an almost commensurate increase in molecularly defined, readily diagnosable, but nonetheless poorly understood and untreatable, diseases. This meeting will examine the current and future bottlenecks to gene discovery, disease modeling, and therapeutic approaches and suggest strategies to enable progress in this regard. Ultimately, successful deployment of precision medicine for rare diseases will inform such approaches more broadly.

Keystone Symposia: Leveraging Genomic Diversity to Promote Animal and Human Health

Genomic variation is a driving force of animal and human health, and susceptibility to disease. Yet our knowledge rarely spans human ethnic genomic diversity and genomic variation between animal breeds, limiting their translational impact. This symposium aims to: 1. Highlight translational genomics in humans and animals (clinical medicine and animal breeding for health and productivity) 2. Explore synergies and cross-disciplinary learning 3. Explore opportunities to leverage genomic diversity to push the current boundaries to translation 4. Address translation and affordability in low- and middle-income settings. Large-scale genomics initiatives like Genome England, the US Precision Medicine initiative, and the Human Heredity and Health in African Consortium, are providing extraordinarily large data sets to explore useful genotype-phenotype connections. Equivalent initiatives for animal data are starting. This meeting will explore the translation of genomic research in animals and humans; high and low-resourced environments; ethnic diversity; and cultural context. Identifying common strands in animal and human health opens up opportunities for repurposing of ideas and applications, and for finding innovative solutions for translational genomics through cross-boundary communities of practice.

The course will introduce state-of-the art modeling and experimental tools for discovering and analyzing species interactions in microbial communities, with a particular focus on metabolic dependencies.                                           The course will run over a period of four days, each day divided between a morning session with theory and an afternoon session with practical work. In the morning session, a leading expert in the field presents background information, key concepts and computational/experimental methods. This lecture module is followed by a round-table discussion chaired by the speaker. The concepts are then introduced in more detail by the speaker or by one of the organizing instructors. Afternoon sessions, supervised by the instructors and teaching assistants, are dedicated to extensive practical work in wet as well as dry lab.
