EMBL Course: High-Accuracy Correlated Light and Electron Microscopy: Applications at Room Temperature and in cryo

Abstract:
This course will teach theory and practice for high-accuracy correlative light and electron microscopy. Students will learn high-accuracy CLEM on resin embedded samples using SEM approaches (Peddie et al. 2014) as well as using TEM according to the procedures developed in the Briggs group at EMBL (Kukulski et al. 2011). The course will also cover the extension of this approach to cryo-EM specimens (Schorb and Briggs 2014), and more recently developed equipment and work flows for cryo-CLEM.

EMBL Course: Brillouin Microscopy: Emerging Tool for Probing Mechanical Properties of Living Cells

Abstract:
Brillouin microscopy is an emerging optical imaging and spectroscopy technique which allows to probe mechanical properties of living cells using light. Mechanical forces have recently become important in cell, tissue and developmental biology as they are thought to regulate a number of different properties and behaviours. Brillouin microscopy offers the ability to measure these mechanical properties in a non-invasive way with high spatial and temporal resolution in living cells, and also gives access to internal structures, i.e. it is not restricted to surface measurements, like many other techniques (e.g. atomic force microscopy). By a mix of lectures and hands-on classes this course aims at bringing this emerging technique closer to the life scientists.

Keystone Symposia - Cryo-EM from Cells to Molecules: Multi-Scale Visualization of Biological Systems

Abstract:
Fast-evolving cryo-EM visualization techniques have recently started to illuminate the inner workings of complicated and dynamic biological assemblies with a level of detail we have never seen before. Cryo-EM is now the state-of-the-art methodology for taking snapshots of biological systems from low to high resolution and with various levels of complexities, from small macromolecular complexes all the way to subcellular organizations. This symposium will showcase cryo-EM breakthroughs in addressing crucial problems in cell biology and will further highlight cryo-EM imaging as a platform for the integration of data from orthogonal approaches. With talks on a variety of biological specimens, procedures and types of biological questions, the meeting aims to attract a very diverse audience from both academia and industry with interest in the method, and beyond the traditional cryo-EM specialist. The symposium will focus on four themes: 1) Single-particle cryo-EM with attention to the characterization of the conformational ensemble adopted by macromolecular complexes and aiming to delineate their mechanism of action; 2) Cryo-electron tomography (cryo-ET) and correlative light electron microscopy (CLEM) to visualize cellular organizations and their dynamics; 3) The emergence of cryo-EM as tool for drug discovery and translational research through both high-resolution structure determination and characterization of cellular phenotypes; and 4) Recent developments in sample preparation and image acquisition. Furthermore, the symposium will include workshops in specimen preparation and evaluation of suitability for cryo-EM, modeling of structures using cryo-EM data, and how to set up a cryo-EM lab.

EMBL Course: Fundamentals of Widefield and Confocal Microscopy and Imaging

Abstract:
Light microscopy is the most widely used form of microscopy for life science research. The correct handling of the microscope and optimising image quality (e.g. contrast, resolution) as well as experimental design according to different applications are crucial to obtain the best results. This course will provide the participants with an overview about the basics of microscopy (light and optics, contrast techniques and imaging). The confocal part will include multicolor, 3D- and time lapse imaging as well as spectral imaging, photobleaching (FRAP, FLIP, FLAP) and photoactivation techniques. Basic image analysis tools including measurement functions, colocalisation and spectral unmixing as well as 3D/4D image processing will be covered, both in theory and practice. The course will combine lectures with intense daily hands-on practices followed by discussions.

Abstract:
This six day advanced course on fluorescence super-resolution microscopy will cover the theory and practical application of current super-resolution microscopy techniques to biological questions. The participants will have sufficient time to use current super-resolution microscopes for Localisation Microscopy (e.g. dSTORM /GSDIM) and STED Microscopy on a variety of fixed and living samples prepared during the course. Invited guest researchers together with experts from the EMBL and Leica Microsystems will ensure that all aspects are covered in depth.

Abstract:
Solving structures of large macromolecular assemblies to gain fundamental insights into biological mechanisms remains a major challenge in structural biology. Cryo-EM combined with image processing is currently undergoing what has been referred to as a “resolution revolution”, making it one of the most powerful techniques to address this challenge. It also has unique capabilities to obtain structural information in situ within complex environments, and to deal with heterogeneous samples. The methods are complex and under constant development, so in depth, advanced training of new users is essential to ensure future users can adapt new technologies to expand into new areas and solve new biological problems. This course provides the theoretical and practical training covering the full breadth of cryo-electron microscopy (cryo-EM) techniques from sample preparation to data collection to image processing.
