This meeting will emphasize the areas of cryo-EM that go beyond what has become the low-hanging fruit of determining structures of stable proteins with single particle cryo-EM. It will look at methods and approaches used for specimen preparation, imaging, and image analysis to allow cryo-EM to solve other problems.

EMBL Course: Fundamentals of Widefield and Confocal Microscopy and Imaging

Abstract:
Light microscopy is the most widely used form of microscopy for life science research. The correct handling of the microscope and optimising image quality (e.g. contrast, resolution) as well as experimental design according to different applications are crucial to obtain the best results. This course will provide the participants with an overview about the basics of microscopy (light and optics, contrast techniques and imaging). The confocal part will include multicolor, 3D- and time lapse imaging as well as spectral imaging, photobleaching (FRAP, FLIP, FLAP) and photoactivation techniques. Basic image analysis tools including measurement functions, colocalisation and spectral unmixing as well as 3D/4D image processing will be covered, both in theory and practice. The course will combine lectures with intense daily hands-on practices followed by discussions.

EMBL Course: Super Resolution Microscopy

Abstract:
This six day advanced course on fluorescence super-resolution microscopy will cover the theory and practical application of current super-resolution microscopy techniques to biological questions. The participants will have sufficient time to use current super-resolution microscopes for Localisation Microscopy (e.g. dSTORM /GSDIM) and STED Microscopy on a variety of fixed and living samples prepared during the course. Invited guest researchers together with experts from the EMBL and Leica Microsystems will ensure that all aspects are covered in depth.

Abstract:
Solving structures of large macromolecular assemblies to gain fundamental insights into biological mechanisms remains a major challenge in structural biology. Cryo-EM combined with image processing is currently undergoing what has been referred to as a “resolution revolution”, making it one of the most powerful techniques to address this challenge. It also has unique capabilities to obtain structural information in situ within complex environments, and to deal with heterogeneous samples. The methods are complex and under constant development, so in depth, advanced training of new users is essential to ensure future users can adapt new technologies to expand into new areas and solve new biological problems. This course provides the theoretical and practical training covering the full breadth of cryo-electron microscopy (cryo-EM) techniques from sample preparation to data collection to image processing.

CISCEM 2018 — 4th International Conference on In Situ and Correlative Electron Microscopy

Abstract:
CISCEM aims at bringing together an interdisciplinary group of scientists from the fields of biology, materials science, chemistry, and physics, to discuss future directions of in-situ electron microscopy research. Topics will include nanoscale studies of biological samples, and functional materials under realistic or near realistic conditions, for example, in gaseous environments, at elevated temperatures, and in liquid. It will be discussed how dynamical processes can be studied by including the time domain in electron microscopy, while taking into account the electron beam effects. CISCEM is also open to other in-situ techniques, such as light microscopy, X-ray, near field or scanning probe microscopy, with the view to stimulate fruitful discussions on multi-scale and correlative approaches.
